Pharmacology
Cannabidihexol (CBDH) is the hexyl-chain homologue of cannabidiol, identified and isolated in 2020 by Linciano, Citti and the Cannazza team in the medical cannabis variety FM2. It fills the gap between the pentyl homologue, cannabidiol, and the heptyl homologue, cannabidiphorol. Isolated by semi-preparative liquid chromatography in a quantity of the order of a tenth of a milligram, its identity was confirmed by comparison with a synthetic standard. In mice, CBDH significantly reduces the late phase of the nociceptive response in the formalin test at doses of 1 and 2 mg/kg, the higher doses of 3 and 5 mg/kg remaining without effect. No affinity for the CB1 or CB2 receptors has been measured. The same study identified Δ9-THCH in this extract at about 7 µg/g without managing to isolate it, unlike CBDH: subsequent mentions of an isolation of THCH are therefore inaccurate.

Biosynthetic pathway
Biosynthetic pathway: CBDH belongs to the long-chain homologue series of cannabidiol, in which the olivetolic acid-type polyketide precursor bears six carbons instead of five. Prenylation and then enzymatic cyclisation lead to cannabidihexolic acid, whose decarboxylation releases CBDH. Its abundance in the plant is very low: the quantity of pure compound obtained on isolation was of the order of a tenth of a milligram.
Legal framework
France
CBDH is not listed by name on the French narcotics list. As a homologue of cannabidiol, it does not share the benzo[c]chromene core of the THC-type derivatives covered by the generic clause of the ANSM Decision of 22 May 2024. Its presence in trace amounts in an extract falls under the framework applicable to hemp.
European Union
Not listed in the European Union narcotics schedules. Concentrated preparations intended for food fall under Regulation (EU) 2015/2283 on novel foods (Novel Food).

Phytocannabinoïde de type cannabidiol, homologue à chaîne hexyle (C6) du CBD. Sa chaîne latérale à six carbones comble l'intervalle entre l'homologue pentyle (le cannabidiol lui-même) et l'homologue heptyle (le cannabidiphorol).

Toxicology and risks
Aucune étude de sécurité humaine n'est disponible. Chez la souris, le CBDH réduit significativement la phase tardive du comportement nociceptif induit par la formaline aux doses de 1 et 2 mg/kg par voie intrapéritonéale, tandis que les doses de 3 et 5 mg/kg sont restées sans effet. Cette relation dose-réponse non monotone reste inexpliquée. Aucune dose humaine caractérisée ne peut être déduite de ces données animales.
Detection and analysis
Le CBDH se détecte par chromatographie liquide ultra-haute performance couplée à la spectrométrie de masse haute résolution (UHPLC-HESI-Orbitrap), technique nécessaire pour le distinguer des autres homologues du cannabidiol dont il ne diffère que par la longueur de chaîne. Son identité a été confirmée par comparaison avec les données spectroscopiques d'un standard de synthèse.
