Pharmacology
Cannabidiol monomethyl ether (CBDM) is a minor cannabinoid of cannabis, structurally identical to cannabidiol except that one of its two phenolic hydroxyls bears a methyl group. Its weak cannabimimetic activity was established in 1986 in a Δ1-THC discrimination test carried out in the rat and the pigeon: at doses of 3 and 10 mg/kg, CBDM produced only vehicle-type responses, and only a dose of 17.5 mg/kg in the pigeon induced 30% of THC-type responses, with an associated depressant effect. Its main interest is mechanistic: metabolism work in the guinea pig showed that liver microsomes convert it into cannabielsoin monomethyl ether by way of a 1S,2R epoxide, under the action of the cytochrome P450 monooxygenase system. This observation helped to elucidate the formation of cannabielsoin-type metabolites from cannabidiol itself.

Biosynthetic pathway
Biosynthetic pathway: CBDM derives from cannabidiol by methylation of one of its phenolic hydroxyls. This simple modification of a single functional group changes the reactivity of the molecule, in particular its metabolic fate, without altering the terpenophenolic scaffold of CBD.
Legal framework
France
CBDM is not listed by name on the French narcotics schedule. A methylated derivative of cannabidiol, it does not share the benzo[c]chromene ring of the THC-type derivatives covered by the generic clause of the ANSM Decision of 22 May 2024. Its presence in trace amounts in an extract falls under the framework applicable to hemp.
European Union
Not listed in the European Union narcotics schedules. Concentrated preparations intended for food use fall under Regulation (EU) 2015/2283 on novel foods (Novel Food).

Cannabinoïde de type cannabidiol dont l'un des deux hydroxyles phénoliques du noyau résorcinol est masqué par un groupement méthyle. Cette méthylation partielle le distingue du cannabidiol, dont les deux hydroxyles sont libres, et du diméthyléther de cannabidiol, où les deux sont méthylés.

Aucune donnée pharmacocinétique humaine n'est disponible pour le CBDM. Les administrations documentées sont intrapéritonéales chez le rat, le pigeon et le cobaye.
Metabolism
Le métabolisme oxydatif du CBDM a été étudié chez le cobaye. Sur microsomes hépatiques, il est transformé en monométhyléther de cannabielsoïne (CBEM), réaction attribuée au système monooxygénase à cytochrome P450. Le mécanisme passe par un époxyde 1S,2R, rapidement converti en métabolite de type elsoïne, exactement comme la formation de la cannabielsoïne à partir du cannabidiol. Cette parenté fait du CBDM un outil ayant servi à élucider le mécanisme de formation des métabolites de type cannabielsoïne du CBD.
Toxicology and risks
Aucune étude de toxicologie humaine dédiée au CBDM n'est publiée, et aucun événement indésirable ne lui est attribué. Sa très faible activité cannabimimétique, mesurée chez deux espèces, suggère l'absence d'effet de type THC aux doses étudiées.
Detection and analysis
Le CBDM se caractérise par chromatographie en phase gazeuse couplée à la spectrométrie de masse, technique employée dans les travaux d'identification de ses métabolites. Sa masse le distingue du cannabidiol, dont il ne diffère que par un groupe méthyle.
