Pharmacology
Cannabidiorcol (CBD-C1, also referenced as O-1821) is the methyl-chain homologue of cannabidiol: the resorcinol skeleton linked to the menthadiene ring is preserved, only the side chain goes from five carbons to a single one. It was identified in hashish in 1972 by Vree and colleagues, alongside the methylated homologues of THC and CBN, and remains a trace constituent of Cannabis sativa. Its affinity for CB1 and CB2 is low, which accords with the structure-activity relationship of the classical cannabinoids, where an overly short side chain strongly degrades receptor recognition. The only activity measured at an identified target comes from the screen by Qin et al. (J Neurosci, 2008): in a calcium mobilisation assay in cells expressing rat TRPV2, the compound produces roughly 93% of the response of the reference agonist 2-APB at the highest concentration tested, that is, an efficacy comparable to that of Δ9-THC, without the authors being able to establish an EC50; it was by contrast practically inactive at TRPA1 and at TRPV1. Beyond this point, the literature is nearly empty: no pharmacokinetics, no characterised metabolism, no toxicity study, and human data are entirely lacking. The hypothesis of an anti-inflammatory effect rests on what is known of TRPV2 in general and not on work devoted to this molecule; the same channel is moreover implicated in tumour proliferation pathways described in experimental oncology, which calls for caution. As things stand, cannabidiorcol belongs to the analytical and chemotaxonomic repertoire of hemp, not to any documented use.

Biosynthetic pathway
The methyl-chain (C1) homologue of cannabidiol. The accepted pathway follows that of the hemp cannabinoids with a shortened polyketide precursor: instead of olivetolic acid derived from hexanoyl-CoA, an acetyl-CoA starter leads to an orcinol-type core (orsellinic acid), then to cannabigerorcinic acid, which CBDA synthase converts into cannabidiorcolic acid; decarboxylation yields cannabidiorcol. This sequence remains a reconstruction by analogy, no enzymology dedicated to the C1 series having been published.
Legal framework
France
Cannabidiorcol is listed by name on no French list of narcotic drugs or psychotropic substances. It is not a tetrahydrocannabinol: the generic clause of annex IV of the decree of 22 February 1990, which classifies "Tetrahydrocannabinols, their esters, ethers, salts and the salts of the aforementioned derivatives", therefore does not capture it. The molecule is unscheduled as such. The raw material, on the other hand, remains regulated, the plant and its resin falling under the narcotics regime, subject to the industrial derogation applicable to cannabis varieties whose Δ9-THC content does not exceed 0.3%.
European Union
No scheduling at European Union level: cannabidiorcol does not appear in the schedules of the United Nations conventions of 1961 and 1971 and no European instrument covers it by name. Control rests on national laws, which generally target cannabis and Δ9-THC rather than the minor cannabinoids. For food use, a cannabinoid isolate falls within the scope of Regulation (EU) 2015/2283 on novel foods and requires prior authorisation.

Cannabinoïde de type résorcinol, squelette cannabidiol : un noyau résorcinol relié à un cycle terpénique menthadiénique, les deux cycles restant ouverts, sans pont pyranique. Se distingue du CBD par sa chaîne latérale, un simple méthyle (C1) au lieu du pentyle (C5), ce qui en fait le plus court des homologues de la série CBD.
Pharmacokinetics
Aucune donnée de pharmacocinétique n'est publiée pour le cannabidiorcol, ni chez l'animal ni chez l'humain : absorption, distribution, demi-vie et biodisponibilité restent inconnues. Sa présence dans la plante se limitant à des traces, l'exposition attendue via un produit de chanvre courant est très faible.
Metabolism
Aucune voie métabolique n'a été caractérisée pour le cannabidiorcol : ni enzyme impliquée (CYP, UGT) ni métabolite identifié. Par analogie avec le cannabidiol, une hydroxylation microsomale suivie d'une conjugaison est plausible, mais rien ne l'a vérifiée expérimentalement pour cet homologue.
Toxicology and risks
Le profil toxicologique n'est pas établi : aucune étude de toxicité, aucun essai clinique et aucune série de cas ne concernent spécifiquement le cannabidiorcol, et les données humaines manquent. La seule réserve mécanistique documentée est indirecte : le TRPV2, sa cible principale, intervient dans des voies de prolifération tumorale décrites en cancérologie expérimentale, ce qui interdit de déduire un bénéfice anti-inflammatoire de la seule activation du canal. En pratique, les quantités présentes dans le chanvre restent trop faibles pour qu'une exposition significative soit attendue.
Detection and analysis
Le composé a été caractérisé dès l'origine par chromatographie en phase gazeuse couplée à la spectrométrie de masse, la chaîne méthyle abaissant la masse moléculaire de 56 unités par rapport au CBD, ce qui rend l'homologue reconnaissable. Il figure aujourd'hui parmi les analytes de méthodes multi-cannabinoïdes validées : une méthode HPLC-MS/MS appliquée à la salive quantifie treize cannabinoïdes dont le CBD-C1. Un étalon de référence dédié reste nécessaire pour le séparer sans ambiguïté des autres homologues de chaîne.
