Pharmacology

Cannabimovone is a minor phytocannabinoid with a rearranged terpene skeleton, isolated in 2010 from a non-psychotropic fibre hemp variety and described as a presumed derivative of cannabidiol. The menthane ring of CBD is contracted there into a cyclopentane, a configuration known as abeo-menthane which sets it apart from THC as from CBD. Iannotti and colleagues (Molecules, 2020) identify it as a selective agonist of PPARγ, active in the low micromolar range in a luciferase assay: the molecule induces the expression of PPARγ target genes involved in the differentiation of adipocytes and prevents, in cells, the impairment of insulin signalling caused by palmitate, which led the authors to present it as a lead in disorders linked to insulin resistance. Dennis and colleagues (Journal of Organic Chemistry, 2022) further report that cannabimovone and its 3' epimer are, along with cannabielsoin, among the minor cannabinoids that reduce the production of pro-inflammatory markers in BV2 microglia stimulated by lipopolysaccharide. No CB1 or CB2 affinity constant is published and no human data exist.

Biosynthetic pathway

The molecule derives from cannabidiol by a rearrangement of the terpene fragment: the six-carbon menthane ring of CBD is contracted into a cyclopentane, which the nomenclature denotes abeo-menthane. The pentylated resorcinol core, common to all phytocannabinoids, is retained, while the terpene fragment now carries a secondary alcohol, an isopropenyl group and a ketone function. No enzyme has been associated with this transformation, which remains described as a presumed conversion of cannabidiol.

Phytocannabinoïde de type abéo-menthane : noyau résorcinol pentylé caractéristique des cannabinoïdes, associé à un fragment terpénique dont le cycle à six carbones a été contracté en cyclopentane portant un alcool secondaire, un groupe isopropényle et une cétone méthylique. Ce squelette réarrangé le distingue des types benzo[c]chromène (THC), résorcinol ouvert (CBD) et dibenzofurane (cannabielsoïne).

Aucune donnée d'absorption, de distribution, de biodisponibilité, de Tmax ou de demi-vie n'a été publiée, ni chez l'animal ni chez l'humain. Les travaux disponibles sont des essais sur cellules et des calculs d'amarrage moléculaire, qui ne renseignent pas sur le devenir de la molécule dans l'organisme.

Metabolism

Aucune voie métabolique n'a été décrite et aucun métabolite n'a été identifié. L'implication éventuelle des cytochromes P450 ou des enzymes de conjugaison n'a jamais été étudiée pour ce composé. La relation inverse est en revanche documentée du côté de la plante, où le cannabimovone est présenté comme issu du cannabidiol.

Toxicology and risks

Aucune étude de toxicité expérimentale, aucun essai de génotoxicité et aucun cas d'exposition humaine ne sont publiés. Les seuls résultats biologiques disponibles proviennent de lignées cellulaires, adipocytes et microglie, où la molécule a été employée à des concentrations micromolaires sans que la cytotoxicité fasse l'objet d'une étude dédiée. Le profil de risque chez l'humain reste inconnu.

Le composé est documenté par les données spectrales de son isolement et de sa synthèse totale, résonance magnétique nucléaire et spectrométrie de masse haute résolution, consultables dans les bases publiques. Aucune méthode validée de recherche en matrice biologique n'a été publiée, et les tests de dépistage courants ciblent les métabolites du THC, pas ce composé. En analyse de matière végétale, il relève des méthodes générales de profilage des cannabinoïdes mineurs par chromatographie couplée à la spectrométrie de masse.
