Pharmacology

Cannabiripsol is a minor cannabinoid of Cannabis sativa, described in 1979 by Boeren, ElSohly and Turner as a 9,10-dihydroxy-hexahydrocannabinol of (6aR,9S,10S,10aR) configuration, isolated from a South African variant and confirmed by synthesis. It retains the benzo[c]chromene nucleus of THC but its terpenic ring is saturated and bears a vicinal diol, which makes it one of the most polar phytocannabinoids known. No measured affinity for CB1 or CB2 has been published: the compound was included in the binding assays and behavioural tests of Radwan and colleagues (Journal of Natural Products, 2015) without any result of its own being reported for it. The CAS numbers 99623-72-0 and 99623-73-1, often associated with cannabiripsol and its propyl homologue in the secondary literature, are probably erroneous: ElSohly and Slade (2005) trace them to a structure incorrectly drawn in a 1985 publication. The identifier retained here is the one that resolves to the correct structure in PubChem.

Biosynthetic pathway

The molecule belongs to the tetrahydrocannabinol lineage: a benzo[c]chromene skeleton arising from cannabigerolic acid by the usual hemp pathway, followed by saturation of the double bond of the terpenic ring and the installation of a vicinal diol at positions 9 and 10. This heavily oxygenated motif suggests an oxidation of the cannabinoid skeleton, in the plant or during the preparation of extracts; no enzyme has been associated with this transformation, described here by chemical class only.
