Pharmacology
CUMYL-BC-HPMEGACLONE-221, also called NBMEGACLONE, is a synthetic cannabinoid of the gamma-carbolinone class, bearing a cumyl group and a norbornylmethyl substituent. In the study by Pulver et al. (2023), the six derivatives compared are full CB1 agonists, and the three norbornylmethyl derivatives have affinities between 0.25 and 1.87 nM, although the value specific to this compound is not given in the abstract consulted. Human metabolism was described from 4 authentic urine samples, which yielded 9 phase I metabolites and two markers of consumption (Giorgetti et al., 2025). In the sources consulted, no clinical study, no human toxicology data and no pharmacokinetic data appear. Its French legal status remains evolving: it is not named in the ANSM list, and the generic definition that could cover it does not settle its case.

History
- Source
- Entirely synthetic molecule, absent from the plant. It has been identified in herbal blends onto which it was applied, and belongs to a group of three related compounds carrying a norbornylmethyl substituent (Pulver et al., 2021).
Origin (pharmaceutical research → illicit market)
No known biosynthetic pathway: a chemically synthesised molecule, with no known natural equivalent, belonging to the class of cumylated gamma-carbolinones.
Legal framework
- European Union
No harmonised European Union control measure was found for this molecule in the sources consulted. It does not appear in the schedules of the 1971 Convention as reproduced in the INCB Green List 2026. Control therefore falls under the national legislation of the Member States, which are not detailed here.
- France
This compound is not named in the consolidated list of substances classed as narcotics published by ANSM, updated 16 February 2026. The generic text covering “3-carboxamide indole derivatives” could apply to it: the nitrogen of the carboxamide bridge is part of a ring, a condition met here. However, the condition on the substituent of the indole nitrogen, “cycloalkylmethyl” or equivalent, is not settled by the text for a bridged bicyclic substituent such as norbornylmethyl. As this cannot be decided with certainty, the status is treated as evolving. The neighbouring compound CUMYL-CH-MEGACLONE, with a cyclohexylmethyl ring, is by contrast named by a decision of 22 May 2024.
