Pharmacology
CUMYL-CB-MEGACLONE is a full agonist of the CB1 receptor and the cyclobutylmethyl analogue of CUMYL-PEGACLONE, built on a gamma-carbolinone core. Pulver et al. (2023) report, for their series of three cyclobutylmethyl SCRAs, Ki values of 0.65 to 29.4 nM and EC50 values of 40.1 to 483 nM, with no isolated constant for this compound in the abstract; Giorgetti et al. (2025) state that it has higher affinity and potency than CUMYL-CBMICA and CUMYL-CBMINACA. The parent molecule was not found in authentic urine, which yielded 15 phase I metabolites: a monohydroxylation of the cyclobutylmethyl chain serves as a marker. No human pharmacokinetic, clinical or toxicological study was identified in the sources consulted. In France, it is not named on the list of narcotics of February 2026 and may fall under a generic clause; internationally, it does not appear in the INCB Green List 2026.

History
- Source
- A fully synthetic molecule, from the clandestine synthesis of SCRAs (Giorgetti et al., 2025). It belongs to the wave of cyclobutylmethyl SCRAs that appeared around 2019-2020. Pulver et al. (2023) state that this side chain made it possible to bypass the German legislation in force at the time.
Origin (pharmaceutical research → illicit market)
No known biosynthetic pathway: a fully synthetic molecule, obtained by clandestine chemical synthesis on a gamma-carbolinone core. Only the class of synthesis is described here.
Legal framework
- European Union
No harmonised control measure at European Union level is documented in the sources consulted: control therefore falls under national legislation. In Germany, the NpSG law was extended in 2019 to the gamma-carbolinone core, then amended in 2020 to place all cyclobutylmethyl SCRAs under control (Giorgetti et al., 2025; Pulver et al., 2023).
- France
Not named in the consolidated list of narcotics updated on 16 February 2026 by ANSM, which does however name several gamma-carbolinones: Cumyl-pegaclone (decision of 30 August 2022), 5F-Cumyl-pegaclone and CUMYL-CH-MEGACLONE (decision of 22 May 2024). It could fall under the generic definition of 3-carboxamide indole derivatives (annex IV). On a literal reading, its indole bears a cycloalkylmethyl group on the nitrogen, and the nitrogen of the carboxamide bridge carries a cumyl group while belonging to a ring. The attachment nonetheless remains uncertain: the carbonyl of the carboxamide is part of the lactam ring fused to the indole core, a case the definition does not expressly provide for. In case of doubt, caution calls for treating it as a narcotic.
