Pharmacology

CUMYL-CH-MEGACLONE is a synthetic agonist of the cannabinoid receptors, of the γ-carbolin-1-one family, and the cyclohexylmethyl analogue of CUMYL-PEGACLONE. Haschimi et al. (2021) measure at hCB1 a Ki of 1.01 nM, an affinity about 2.5 times higher than that of JWH-018, and an EC50 of 1.22 nM, with high efficacy (Emax of 143.4% above the receptor's constitutive activity). The authors suggest that it more closely resembles CUMYL-PEGACLONE pharmacologically. First identified in Hungary in December 2018 in a herbal blend, it was later found in samples from intoxicated patients. The file remains thin: no clinical study and no assessment of abuse potential in humans was found in the sources consulted, pharmacokinetics is documented only through a few blood samples, and the molecule is not listed in the INCB 2026 Green List, although it is scheduled as a narcotic in France.

History

Source
Fully synthetic molecule, with no known plant origin. It was identified in a herbal blend in which it was the main active ingredient. Its γ-carbolinone nucleus places it in the same chemical family as CUMYL-PEGACLONE (Haschimi et al., 2021).

Origin (pharmaceutical research → illicit market)

No known biosynthetic pathway: a synthetic molecule of the γ-carbolin-1-one class, with no known link to hemp or other plants.
