Pharmacology

Cumyl-NBMINACA is a synthetic cannabinoid of the indazole-3-carboxamide family, a full agonist at the CB1 cannabinoid receptor. Pulver et al. (2023) report, for the norbornylmethyl series, an affinity constant Ki between 0.25 and 1.87 nM, the indazole being the most affine derivative, with an EC50 from 1.78 to 169 nM for the same series. Giorgetti et al. (2025) described its phase I metabolism from two authentic urine samples, in which 13 phase I metabolites were identified, and proposed urinary biomarkers based on the norbornylmethyl chain. Its legal status in France remains to be settled: it is not named on the ANSM list, and whether it meets the generic definition of indazole-3-carboxamides depends on how the cycloalkylmethyl substituent is interpreted. No clinical study and no controlled human toxicology data are published in the sources consulted.

History

Source
Synthetic molecule: the sources consulted class it among synthetic cannabinoids, with no plant origin reported in those sources. Purified standards of Cumyl-NBMINACA were isolated from herbal blend material within the European ADEBAR plus project (Giorgetti et al., 2025). Pulver et al. (2023) report that the cyclobutylmethyl and norbornylmethyl side chains allowed these substances to circumvent German legislation, and that the market life cycle of these substances was less than one year.

Origin (pharmaceutical research → illicit market)

No biosynthetic pathway: Cumyl-NBMINACA is a synthetic molecule, chemically classed among the indazole-3-carboxamides, with no known link to a plant or a living organism.
