- Les deux stéréoisomères ont été décrits en 1984 par Srebnik, Lander, Breuer et Mechoulam, qui les obtiennent par isomérisation du Δ9-THC en milieu basique puis les séparent par chromatographie. La molécule est restée un objet de laboratoire pendant plus de trente ans, avant de réapparaître vers 2020 dans les produits issus de la conversion du CBD, d'abord aux États-Unis.

In plain terms
Δ10-THC is a chemical cousin of THC: the same molecular formula, with the double bond placed elsewhere on the skeleton. Hemp makes practically none of it; it comes from the chemical transformation of CBD or of Δ9-THC in the laboratory. Test-tube assays show that it binds to the cannabinoid receptor of the brain without activating it, and no study has been carried out in humans.

Pharmacology
Δ10-THC shares the dibenzopyran skeleton of Δ9-THC, with a double bond moved to position 10 and an additional asymmetric centre at C9, which gives rise to cis and trans epimers. In radioligand displacement at the CB1 receptor, both epimers bind measurably (IC50 of 29.1 nM for the trans and of 294.2 nM for the cis), but remain markedly less avid than Δ9-THC (2.1 nM in the same study). The decisive point is functional: neither epimer produced an agonist effect up to the micromolar concentrations tested, and both behave instead as antagonists of CP55,940, a property that had never been described for another isomer of THC. This result agrees with the animal data considered by the WHO Expert Committee, in which Δ10-THC did not substitute for Δ9-THC in pigeons trained to discriminate that molecule. Nothing is documented on CB2, TRPV1, GPR55, PPARγ or 5-HT1A, and human data are entirely lacking: no pharmacokinetics, no clinical trial, no study of subjective effects.

Route of preparation (chemical process)
No known enzymatic pathway: hemp does not produce this isomer. Δ10-THC is obtained by isomerisation of Δ9-THC under basic conditions, or arises as a by-product of the acid cyclisation of cannabidiol, the reaction used industrially to manufacture Δ8-THC.
Legal framework
France
In France, Δ10-THC is not listed by name by the ANSM. It falls under the generic clause of annex IV of the decree of 22 February 1990, which classifies as narcotics "Tetrahydrocannabinols, their esters, ethers, salts as well as the salts of the aforementioned derivatives": Δ10-THC being a tetrahydrocannabinol, it falls under that family entry, and not under an individual listing. Production, possession, transfer and use are therefore prohibited, and a consumer product containing it is unlawful whatever its CBD content may be in other respects.
European Union
Internationally, Δ10-THC is among the six isomers of tetrahydrocannabinol listed by name in Schedule I of the 1971 Convention on Psychotropic Substances, under the designation 6a,7,8,9-tetrahydro-6,6,9-trimethyl-3-pentyl-6H-dibenzo[b,d]pyran-1-ol. The European Union does not maintain a single narcotics list: each member state transposes the convention into its national law, most often through a generic entry covering tetrahydrocannabinols, and no control measure specific to the Union covers this isomer in particular. The applicable regime therefore varies from one country to another, even though the United Nations listing results in practice in narcotic status in most member states.

Pharmacokinetics
Aucune étude de pharmacocinétique humaine n'a été publiée. Le comité d'experts de l'OMS relève que les données propres aux isomères du THC sont rares et raisonne par analogie : comme les autres tétrahydrocannabinols, le Δ10-THC est très lipophile, donc absorbé et distribué vers le cerveau par voie orale, inhalée ou parentérale, avec un métabolisme initial hépatique assuré par les cytochromes P450. Demi-vie, biodisponibilité et volume de distribution restent inconnus chez l'humain.
Metabolism
Le seul jeu de données spécifique provient d'hépatocytes humains : là où le Δ8-THC et le Δ9-THC forment des métabolites monohydroxylés puis carboxylés ainsi que leurs glucuronides, le Δ10-THC suit une voie distincte, dominée par la glucuronidation directe du composé parent et par une monohydroxylation, la formation du métabolite carboxylé restant marginale. Les auteurs soulignent le risque d'erreur d'interprétation en toxicologie clinique et médico-légale, puisque le marqueur habituellement recherché pour les tétrahydrocannabinols est précisément ce métabolite carboxylé.
Toxicology and risks
Le profil toxicologique n'est pas établi. Le comité d'experts de l'OMS indique que le Δ10-THC n'a fait l'objet d'aucune évaluation détaillée de sa toxicité et qu'aucun volontaire n'a reçu la molécule pure. Les signaux disponibles portent sur les produits, pas sur la substance : les centres antipoison des États-Unis ont enregistré plus de cinq mille expositions à des articles vendus comme Δ8-THC, Δ10-THC ou THC-O acétate en 2021 et 2022, avec dépression du système nerveux central, tachycardie et agitation au premier rang, plus d'un tiers d'issues médicales sérieuses et près d'un tiers des cas chez des enfants de moins de six ans ; le Δ8-THC concentrait toutefois 98 % de ces dossiers. S'ajoute un problème de composition : les articles étiquetés Δ10-THC contiennent régulièrement du Δ6a,10a-THC, d'autres isomères et des résidus de réaction non caractérisés.
Detection and analysis
Les isomères se séparent en chromatographie gazeuse couplée à la spectrométrie de masse après dérivation : c'est par cette voie que les deux diastéréoisomères 6aR,9R et 6aR,9S du Δ10-THC ont été identifiés dans des liquides de vapotage saisis, aux côtés du Δ6a,10a-THC. En dépistage urinaire, le Δ10-THC et ses analogues carboxylés réagissent de façon croisée avec les immunodosages cannabinoïdes du commerce, avec une sensibilité qui dépend du réactif et du seuil retenu : un dépistage positif ne permet donc pas de distinguer le Δ10-THC du Δ9-THC, et seule une confirmation chromatographique tranche.
