- Signalé en 2020 par Linciano, Citti, Cannazza et leurs collègues, dans un article de Scientific Reports consacré au cannabidihexol. Les auteurs ont repéré le Δ9-THCH et sa forme acide dans la variété médicinale italienne FM2 par chromatographie liquide couplée à la spectrométrie de masse haute résolution, puis ont confirmé l'attribution en comparant les spectres à un étalon obtenu par synthèse stéréosélective. Le composé n'a jamais été isolé du végétal : les auteurs indiquent expressément que son abondance était trop faible pour en récupérer une fraction pure. La chimie des homologues hexyle du squelette cannabinoïde est bien plus ancienne, puisque Roger Adams avait exploré dès les années 1940 la série des homologues alkyle, dont le parahexyl, isomère de position du Δ9-THCH.

In plain terms
Tetrahydrocannabihexol is a very close cousin of THC: the same molecular framework, with a small carbon tail that is slightly longer. It was spotted in 2020 in an Italian medicinal hemp variety, in quantities so minute that the researchers were never able to extract the least pure sample of it. What this molecule actually does in the body remains unknown: no animal or human study has been published about it.

Pharmacology
Δ9-THCH is the n-hexyl homologue of Δ9-THC: the alkyl chain grafted at position 3 of the benzo[c]chromene nucleus has six carbons instead of five. That position precisely commands anchoring in the lipophilic pocket of the CB1 receptor, and the structure-activity relationship established across the homologous series shows affinity rising from three carbons, peaking around eight carbons and then declining. On that basis, Δ9-THCH should behave as a partial CB1 agonist, with an affinity at least comparable to that of Δ9-THC and situated between the latter and heptyl Δ9-THCP. The status of that statement deserves emphasis: it is a structural deduction, not an experimental result. The 2020 work that reported the molecule carried out on it no binding assay, no functional test and no animal experiment; only the cannabidihexol described in the same article was evaluated in mice. Nothing is established either for TRPV1, GPR55, PPARγ or 5-HT1A. Human data are entirely lacking, and the commercial claims putting its potency at ten or twenty-five times that of THC rest on no scientific publication.

Biosynthetic pathway
The natural pathway remains undetermined. Hemp cannabinoids classically carry a side chain with an odd number of carbons (propyl at C3, pentyl at C5, heptyl at C7), inherited from the fatty-acid precursor that initiates the polyketide pathway. Δ9-THCH is an exception with its six-carbon hexyl chain, and the authors of the 2020 work recall that even-numbered homologues had until then been assumed to be artefacts arising from fungal omega-oxidation of the corresponding homologues. They acknowledge that they have not settled the question of a genuine plant origin versus a microbial transformation, and leave it open.
Legal framework
France
Prohibited in France through a generic clause, without being named. Annex IV of the decree of 22 February 1990 classifies as narcotics "tetrahydrocannabinols, their esters, ethers, salts and the salts of the aforementioned derivatives", a formulation that encompasses the homologues of THC. The ANSM decision of 22 May 2024, which came into application on 3 June 2024, added an explicit structural clause covering "any substance derived from the benzo[c]chromene nucleus", substituted at position 1 by a hydroxyl function, at position 3 by an alkyl chain, at position 6 by one or two alkyl groups and at position 9 by an alkyl group. Δ9-THCH matches that description point by point and appears in none of the exceptions provided for (cannabinol, cannabinolic acid, THCV and THCVA below threshold, tetrahydrocannabinolic acid below threshold). Its status is therefore that of a narcotic captured by a generic clause rather than that of a substance listed by name: production, sale, possession and use are prohibited. Parahexyl, a positional isomer, is for its part named in the same annex.
European Union
No control measure at European Union level names Δ9-THCH. New psychoactive substances fall under the early warning system of the EUDA, formerly the EMCDDA, which monitors several hundred cannabinoids, and effective control remains a national competence: status therefore varies from one Member State to another, several countries having adopted, as France has, generic definitions covering the homologues of THC. Internationally, the 1971 Convention on Psychotropic Substances has listed parahexyl in Schedule I since 1982, but Δ9-THCH, a distinct isomer, is not named there.

Pharmacokinetics
Aucune donnée pharmacocinétique publiée : ni absorption, ni distribution, ni demi-vie n'ont été mesurées, chez l'animal comme chez l'être humain. La lipophilie très élevée conférée par la chaîne hexyle laisse attendre un stockage dans les tissus adipeux et une élimination lente, comme pour les autres cannabinoïdes de cette famille, mais il s'agit d'une inférence structurale et non d'une mesure.
Metabolism
Aucune étude de métabolisme n'a été conduite sur le Δ9-THCH. Par analogie avec le Δ9-THC, une hydroxylation hépatique portée par les cytochromes CYP2C9 et CYP3A4, suivie d'une oxydation en acide carboxylique puis d'une glucuroconjugaison, constitue l'hypothèse la plus attendue, mais aucun métabolite du Δ9-THCH n'a été caractérisé à ce jour.
Toxicology and risks
Le profil toxicologique est inconnu. Aucune étude de toxicité aiguë ou répétée, aucun essai chez l'animal et aucune donnée clinique ne portent sur cette molécule ; les données humaines manquent totalement. Aucun cas d'intoxication spécifiquement attribué au Δ9-THCH n'est documenté dans la littérature médicale, ce qui traduit l'absence d'analyse ciblée bien plus qu'une innocuité démontrée. Les risques généraux associés aux agonistes puissants de CB1 restent à considérer : tachycardie, variations de la pression artérielle, anxiété aiguë, épisodes confusionnels, vomissements. Les produits vendus sous ce nom échappent à tout contrôle de composition, et les analyses européennes de cannabinoïdes semi-synthétiques montrent régulièrement des écarts entre l'étiquette et le contenu réel.
Detection and analysis
L'identification repose sur la chromatographie liquide couplée à la spectrométrie de masse haute résolution, méthode employée lors du signalement initial (ionisation électrospray, analyseur Orbitrap). La difficulté principale tient à l'isomérie : la formule brute C22H32O2 est partagée par le parahexyl et par d'autres cannabinoïdes hexyle, si bien que la masse exacte ne suffit pas et qu'une séparation chromatographique adossée à un étalon de référence devient indispensable. Les immunoessais urinaires courants du cannabis, calibrés sur les métabolites du Δ9-THC, ne sont pas validés pour cette molécule, et les méthodes GC-MS de routine ne la signalent pas spontanément. Aucun métabolite marqueur n'étant décrit, il n'existe pas de méthode de dépistage validée en toxicologie médico-légale.
