Cravatt (Scripps Research) en collaboration avec le groupe de Mario van der Stelt (université de Leyde), dans un article des PNAS consacré au remodelage des réseaux lipidiques cérébraux après blocage aigu des diacylglycérol lipases. Pharmacology

DH376 is a 1,2,3-triazole urea designed as a chemical probe of the endocannabinoid system. It carbamoylates the catalytic serine of diacylglycerol lipases alpha and beta and therefore blocks 2-AG synthesis covalently and irreversibly. Published values place potency at around 6 nM at DAGLα and 3 to 8 nM at DAGLβ, with a pIC50 of 8.2 to 8.9 at the human enzyme in the Guide to Pharmacology database. In the mouse, an intraperitoneal injection blocks brain DAGLα activity with a median effective dose of 5 to 10 mg/kg, inhibition becoming complete around 30 to 50 mg/kg. The seminal work showed that acute blockade reorganises the lipid networks of the brain within a few hours: collapse of 2-AG, accumulation of diacylglycerols, a fall in arachidonic acid and in prostaglandins PGD2 and PGE2, and attenuation of the response to lipopolysaccharide, including anapyrexia. The literature establishes no therapeutic benefit: DH376 has never been evaluated in humans, human data are entirely lacking, and imperfect selectivity with respect to ABHD6 and PLA2G7 remains a limitation acknowledged by the authors themselves, who observed certain behavioural effects even with a control compound inactive at the DAGLs.

No biosynthetic pathway: DH376 forms in no living organism. The molecule is obtained by multi-step organic synthesis, by coupling a chiral 2,5-disubstituted piperidine to a 1,2,3-triazole bearing a bis(4-fluorophenyl)carbinol group, the urea function then ensuring covalent binding to the target enzyme.

Urée de 1,2,3-triazole N2-carbamoylée, associant une pipéridine chirale 2,5-disubstituée à un triazole porteur d'un groupe bis(4-fluorophényl)carbinol. Inhibiteur covalent de sérine hydrolase, sans aucune parenté structurale avec les cannabinoïdes végétaux.

Pharmacokinetics

Les données proviennent uniquement du rongeur. Administrée par voie intrapéritonéale à la souris, la molécule franchit la barrière hémato-encéphalique et inhibe la DAGLα cérébrale avec une dose efficace médiane de 5 à 10 mg/kg, l'inhibition devenant complète vers 30 à 50 mg/kg, et la baisse du 2-AG étant mesurable en deux heures. L'inhibition étant covalente, la durée d'action dépend surtout du renouvellement de la protéine cible : aux faibles doses, l'activité DAGLα se rétablit en quelques heures, ce qui a conduit les auteurs à décrire la DAGLα comme une protéine à demi-vie courte dans le cerveau, alors que la cible secondaire ABHD6 reste inhibée plus longtemps. Aucun paramètre pharmacocinétique humain n'existe.

Metabolism

Aucune étude de métabolisme n'a été publiée. Le mécanisme d'action repose sur le transfert du fragment carbamoyle vers la sérine catalytique de l'enzyme, avec libération du triazole substitué comme groupe partant ; le devenir de ce fragment libéré, ainsi que les voies de dégradation hépatique et les enzymes du cytochrome P450 éventuellement impliquées, ne sont pas documentés.

Toxicology and risks

Aucune étude de toxicologie réglementaire n'a été conduite et la molécule n'a jamais été administrée à l'humain. Chez la souris, les doses employées en recherche sont tolérées sur la durée des expériences, mais la sélectivité imparfaite constitue le principal signal de vigilance : au-delà des diacylglycérol lipases, DH376 inhibe ABHD6 et PLA2G7, et à moindre degré CES1C, ABHD2, la butyrylcholinestérase et LIPE. Les auteurs des travaux sur la reprise alimentaire après jeûne ont d'ailleurs observé un effet comparable avec un composé témoin inactif sur les DAGL, ce qui indique que plusieurs cibles contribuent aux observations. Toute exposition humaine doit être considérée comme un risque non caractérisé.

Detection and analysis

Aucune méthode de dépistage de routine n'existe et les laboratoires de toxicologie médico-légale ne recherchent pas cette molécule, faute de circulation hors du milieu académique. En recherche, l'engagement de la cible se mesure par profilage protéomique fondé sur l'activité, à l'aide de sondes qui marquent les sérine hydrolases restées actives, couplé à la spectrométrie de masse. La fonction alcyne portée par DH376 permet en outre de greffer un fluorophore par chimie click, ce qui a donné la sonde fluorescente DH379 utilisée pour visualiser les diacylglycérol lipases.
