In plain terms
DO34 is not a hemp cannabinoid: it is a tool created in the laboratory to cut off the manufacture of 2-AG, one of the cannabinoids the body produces itself. In the mouse, a single injection is enough to make the level of 2-AG in the brain fall within a few hours. This molecule serves research only, has never been given to humans and enters into no product on sale.

Pharmacology
DO34 is a 1,2,3-triazole urea designed as a chemical probe of the endocannabinoid system. It does not act at the CB1 and CB2 receptors, where its binding is negligible beyond 1 µM, but covalently acylates the catalytic serine of the diacylglycerol lipases alpha and beta, the two enzymes that produce 2-arachidonoylglycerol. The values reported are close to 6 nM at recombinant human DAGL-alpha and 3 to 8 nM at DAGL-beta. In the mouse, an intraperitoneal injection blocks brain DAGL-alpha with an ED50 of 5 to 10 mg/kg and complete inhibition around 30 to 50 mg/kg: the brain level of 2-AG falls by about 80 % within two hours, accompanied by a decrease in anandamide, arachidonic acid and the prostaglandins PGD2 and PGE2, and by an accumulation of the precursor diacylglycerols. In cerebellar and hippocampal slices, the molecule abolishes the forms of synaptic plasticity that depend on 2-AG. The reverse side is documented in rodents: depletion of 2-AG increases anxiety, promotes vulnerability to stress and impairs fear extinction as well as spatial learning at high doses. Selectivity is not absolute, ABHD6, PLA2G7, ABHD2, CES1C and PAFAH2 being affected as well, which makes the use of the inactive control DO53 necessary in experimental protocols. Human data are entirely lacking: no clinical study, no pharmacokinetic profile in humans, no toxicity data.

Origin (research tool)
No biosynthetic pathway. DO34 is a product of total synthesis belonging to the class of 1,2,3-triazole ureas, with no plant precursor and no known enzyme capable of forming it.
Legal framework
France
DO34 is named in no French decree. It is not a tetrahydrocannabinol, so neither is it covered by the generic clause of annex IV of the decree of 22 February 1990, which covers tetrahydrocannabinols, their esters, ethers and salts as well as the salts of these derivatives. The situation is that of a substance not classified as a narcotic, but devoid of any status as a medicine or an ingredient: it circulates solely as a research reagent, under the labelling and safety obligations applicable to laboratory chemicals, and its administration to humans is authorised in no framework.
European Union
No control measure at Union level has been identified. The molecule does not figure among the substances added to the definition of drug in Framework Decision 2004/757/JHA, and it has not been the subject of a notification to the early warning system coordinated by the EUDA, formerly the EMCDDA. It falls under the general regime for chemicals, the REACH and CLP regulations, with use restricted to research.

Urée de 1,2,3-triazole, famille des inhibiteurs covalents de sérine hydrolases. Le squelette associe une pipérazine protégée par un groupement tert-butyloxycarbonyle et portant un benzyle, reliée par une fonction urée à un triazole substitué par un phényle trifluorométhoxylé qui joue le rôle de groupe partant. Aucune parenté structurale avec les cannabinoïdes terpénophénoliques du chanvre.
Pharmacokinetics
Les données disponibles proviennent exclusivement de la souris. Par voie intrapéritonéale, DO34 franchit la barrière hématoencéphalique et engage sa cible dans le cerveau, avec une DE50 de 5 à 10 mg/kg sur la DAGL-alpha et une inhibition complète entre 30 et 50 mg/kg. L'effet est rapide, mesurable dès deux heures, et se maintient jusqu'à huit heures à forte dose, avec une récupération partielle de l'activité enzymatique à vingt-quatre heures. Cette cinétique reflète autant la demi-vie brève de la protéine DAGL-alpha, rapidement renouvelée, que la persistance du composé. Aucun paramètre pharmacocinétique humain n'est publié.
Metabolism
Le mode d'action repose sur une acylation covalente : le fragment pipérazine reste fixé sur la sérine catalytique de l'enzyme tandis que le triazole se détache comme groupe partant, si bien que le retour à une activité normale dépend de la resynthèse de la protéine plutôt que de l'élimination du composé. Le devenir métabolique proprement dit, voies hépatiques et métabolites circulants, n'a pas été caractérisé dans la littérature publiée, ni chez l'animal ni chez l'homme.
Toxicology and risks
Aucune donnée de toxicité humaine. Chez le rongeur, les signaux d'alerte sont neurocomportementaux et découlent de la perte de signalisation 2-AG : anxiété accrue, sensibilité augmentée aux conséquences du stress, extinction de la peur dégradée et apprentissage spatial ralenti aux doses élevées, avec une perte de poids lors d'une administration répétée à 30 mg/kg. L'invalidation génétique de la DAGL-alpha, qui produit un état comparable, s'accompagne d'hypophagie et d'une propension aux crises convulsives, ce qui constitue le principal motif de prudence pour tout inhibiteur pénétrant dans le système nerveux central. La sélectivité incomplète sur d'autres sérine hydrolases ajoute une source d'effets hors cible.
Detection and analysis
Aucune méthode de dépistage de routine n'existe : la molécule n'est pas recherchée en toxicologie clinique ou médico-légale et n'a pas été signalée sur le marché des produits de consommation. En laboratoire, l'identification passe par chromatographie liquide couplée à la spectrométrie de masse, et l'engagement de la cible se mesure par profilage d'activité protéique en compétition, à l'aide de sondes dédiées aux diacylglycérol lipases.
