First described
Isolée, caractérisée et nommée en 1974 à partir des graines de Piper guineense, le poivre de Guinée, d'où son nom. Elle a ensuite été retrouvée dans le poivre long et le poivre noir, et décrite en 2004 comme inhibiteur de l'ACAT dans les fruits de Piper longum. Son intérêt pour le système endocannabinoïde est plus récent : en 2014, l'équipe de Jürg Gertsch à l'Université de Berne l'identifie par criblage à haut contenu comme inhibiteur nanomolaire de la recapture cellulaire de l'anandamide. Une synthèse totale accompagnée d'une étude de relations structure-activité a suivi en 2019.

Guineensine is a piperamide, that is to say an amide alkaloid, present in the seeds of Piper guineense, in long pepper (Piper longum) and in black pepper (Piper nigrum). It therefore belongs to the ordinary diet and is found in any culinary preparation based on pepper, alongside piperine, which is far more abundant.

Guineensine is a molecule of pepper: it is found in black pepper, long pepper and Guinea pepper. It does not come from hemp and has no structural kinship with CBD or THC. In mice, it slows the recovery by cells of the cannabinoids that the body makes itself, which prolongs their action; in human beings, nothing of the kind has been measured.

Pharmacology

Identified in 2014 by high-content screening, guineensine inhibits the cellular reuptake of anandamide with a potency of the order of a few hundred nanomolar, and also blocks that of 2-arachidonoylglycerol. Its originality lies in the fact that it avoids the classical targets: no affinity for CB1 or CB2, no inhibition of FAAH or MAGL, no interaction with the carrier protein FABP5, no inhibition of the serine hydrolases in activity profiling. It acts upstream, on the membrane transport of the endocannabinoids, and indirectly reinforces their signalling. In mice, this produces a complete cannabimimetic picture, combining catalepsy, hypothermia, hypolocomotion and analgesia; catalepsy, analgesia and hypothermia are reversed by rimonabant, which confirms the CB1 relay, whereas hypolocomotion is not. Anti-inflammatory effects were reported in 2017 in mouse models of inflammatory pain, of oedema and of endotoxaemia. What is not established deserves to be stated just as clearly: no clinical study, no pharmacokinetic or safety data in human beings, and an incomplete identification of the transporter targeted, whose very existence is still the subject of discussion.

The piperamide pathway of the genus Piper: the acyl part derives from the phenylpropanoid pathway, which supplies the methylenedioxyphenyl (benzodioxole) motif from cinnamic acid, then undergoes a polyketide chain elongation giving the trienoic skeleton. This chain is then coupled as an amide with the isobutylamine derived from valine. Guineensine differs from piperine only by the length of the chain and by the nature of the amine, piperine carrying a piperidine derived from lysine.

Alcaloïde amide de type pipéramide : N-isobutylamide à longue chaîne triénoïque portant un motif 1,3-benzodioxole. Proche parente de la pipérine, elle n'a aucune parenté avec le squelette terpénophénolique des cannabinoïdes végétaux et se rapproche plutôt, par sa forme allongée et lipophile, des amides d'acides gras du système endocannabinoïde.

Pharmacokinetics

Les données publiées sont limitées. La molécule est très lipophile et non ionisable, et les effets centraux observés chez la souris après administration intrapéritonéale, catalepsie, hypothermie et analgésie, indiquent qu'elle franchit la barrière hémato-encéphalique. Les effets décrits dépendent à la fois de la dose et du temps, avec une action qui se maintient au-delà de l'administration. Aucun paramètre humain n'est documenté : ni biodisponibilité orale, ni demi-vie, ni volume de distribution, ni concentration plasmatique atteinte après consommation alimentaire de poivre.

Metabolism

Le métabolisme de la guinéensine n'a pas été caractérisé. Par analogie avec les autres pipéramides alimentaires, on attend une hydrolyse de la liaison amide libérant l'isobutylamine et l'acide correspondant, ainsi qu'une transformation oxydative du motif méthylènedioxyphényle par les cytochromes P450, avec le risque d'inhibition enzymatique que cette famille chimique comporte. Aucune étude d'identification de métabolites, chez l'animal ou chez l'être humain, n'a été publiée à ce jour.

Toxicology and risks

Aucune étude de toxicité réglementaire n'est disponible pour la molécule isolée. Le seul élément rassurant tient à son statut alimentaire : elle est ingérée de longue date avec le poivre, à des niveaux très inférieurs à ceux employés dans les études animales. Chez la souris, les doses pharmacologiquement actives provoquent une catalepsie marquée, une hypothermie et une réduction nette de l'activité motrice, effets de type cannabimimétique qui traduisent une action centrale réelle. Les données humaines manquent totalement : ni seuil sans effet, ni profil d'effets indésirables, ni interactions médicamenteuses ne sont établis. La prudence s'impose donc pour tout produit qui concentrerait cette molécule au-delà des teneurs présentes dans l'épice.

Detection and analysis

La guinéensine se dose dans les épices par chromatographie liquide couplée à la spectrométrie de masse ou à une détection par barrette de diodes, méthodes employées pour profiler les pipéramides des différentes espèces de Piper. Aucun protocole de dépistage toxicologique ou forensique ne la recherche : elle n'est pas surveillée dans les matrices biologiques et, n'ayant aucune parenté structurale avec le THC, elle n'est pas ciblée par les immunoessais cannabinoïdes utilisés en routine.
