- La molécule a été décrite pour la première fois en février 2001 par Hiroshi Iwamura et ses collègues de Japan Tobacco, dans un article du Journal of Pharmacology and Experimental Therapeutics consacré à sa caractérisation in vitro et in vivo comme ligand sélectif du récepteur cannabinoïde CB2. C'était alors le premier rapport montrant qu'un agoniste inverse sélectif du CB2 possédait un effet anti-inflammatoire chez l'animal. Le composé a ensuite servi de molécule de référence à une étude de relations structure-activité publiée en 2006 par une équipe finlandaise de l'université de Kuopio, qui a exploré une série d'amides aromatiques bâtis sur le même squelette 2-oxoquinoléine.

In plain terms
JTE-907 is a laboratory molecule developed by a Japanese pharmaceutical group. It does not exist in hemp: it was designed to switch off the background activity of the CB2 receptor, one of the two receptors on which cannabinoids act, found mainly in cells of the immune system. In mice it attenuates certain skin inflammations, but it has never been evaluated in humans and is not present in any CBD product.

Pharmacology
JTE-907 is a selective inverse agonist at the CB2 cannabinoid receptor, developed by Japan Tobacco as a research tool. The affinities measured by Iwamura and colleagues in 2001 show a strong species dependence: the Ki is 35.9 nM at human CB2, 1.55 nM at mouse CB2 and 0.38 nM at rat CB2, with selectivity ratios against CB1 of 66, 684 and 2760 respectively. Functionally, the molecule increases cyclic AMP production in cells expressing CB2, the signature of inverse activity rather than simple blockade; [35S]GTPγS assays place it among the full inverse agonists, with an efficacy comparable to SR 144528. In mice, oral administration reduces carrageenan-induced paw oedema, as well as ear swelling provoked by an analogue of 2-arachidonoylglycerol, by dinitrofluorobenzene or by an IgE-dependent skin reaction, results that led CB2 inverse agonists to be proposed as an anti-inflammatory and anti-allergic avenue. That reading calls for several reservations. Later work classified the molecule among the protean ligands, whose effect reverses after repeated administration once the constitutive activity of CB2 has been abolished, and a study on pancreatic islets revealed a stimulatory action coupled to Gq proteins, independent of GPR55 and therefore off-target. No clinical trial has been published and human data are entirely lacking: the molecule remains a laboratory reagent, with no established therapeutic indication and no safety profile in humans.

Origin (research tool)
No known biological pathway: the molecule is not formed in any living organism. It is obtained by medicinal chemistry, in the 2-oxoquinoline carboxamide class, with no relation to the biosynthetic pathway of plant cannabinoids, which proceeds via olivetolic acid and geranyl pyrophosphate.
Legal framework
France
JTE-907 does not appear in any of the annexes of the decree of 22 February 1990 setting the list of substances classified as narcotics, and it is not named in any ANSM decision. The generic clause of annex IV, which covers tetrahydrocannabinols, their esters, ethers and salts as well as the salts of the aforementioned derivatives, does not apply either: the molecule is not a tetrahydrocannabinol and is neither an ester, an ether nor a salt of one, but a 2-oxoquinoline carboxamide of an entirely different structure. Its French status is therefore that of an unscheduled substance, falling under the ordinary law governing laboratory chemicals. This does not mean that it would be authorised for consumption: it holds no marketing authorisation, may not be marketed as a medicine, as a food supplement or as a cosmetic ingredient, and any placing on the market for human use would be unlawful on that basis.
European Union
No European Union instrument places JTE-907 under control: the substance has been the subject of no measure under Framework Decision 2004/757/JHA as amended, nor of a risk assessment by the European Union Drugs Agency, for want of circulation on the new psychoactive substances market. It is listed in no schedule of the international conventions of 1961 and 1971 and has not been examined by the World Health Organization Expert Committee on Drug Dependence. It is, however, registered with the European Chemicals Agency, where the notifications received classify it as acute oral toxicity category 4, with the hazard statement “harmful if swallowed”. National regulations vary, some States attaching synthetic cannabinoid ligands to generic clauses whose scope depends on the wording adopted by each legislator.

Carboxamide de 2-oxo-1,2-dihydroquinoléine, portant un groupement méthoxy en position 7, une chaîne pentyloxy en position 8 et un bras benzodioxol-5-ylméthyle sur la fonction amide. La molécule n'appartient pas à la famille des cannabinoïdes du chanvre : elle ne possède ni squelette terpénophénolique, ni noyau résorcinol à chaîne alkyle, et se range parmi les outils pharmacologiques de synthèse du système endocannabinoïde.
Pharmacokinetics
Les données publiées se limitent à l'animal. La molécule est active par voie orale chez la souris, dans une gamme de doses de l'ordre du milligramme par kilogramme, ce qui suppose une absorption digestive suffisante et une exposition systémique utile. Sa lipophilie calculée est élevée, cohérente avec une bonne diffusion tissulaire. Aucune valeur de biodisponibilité, de demi-vie d'élimination, de volume de distribution ni de liaison aux protéines plasmatiques n'a été publiée, et il n'existe aucune donnée de pharmacocinétique humaine.
Metabolism
Aucune étude de métabolisme n'a été publiée pour cette molécule. Sa structure comporte plusieurs sites classiquement pris en charge par les monooxygénases hépatiques : un éther méthylique, une chaîne pentyloxy susceptible d'être hydroxylée puis raccourcie, un motif méthylènedioxyphényle et une fonction amide hydrolysable. Ces voies sont plausibles au vu de la structure, mais elles n'ont pas été démontrées expérimentalement et aucun métabolite n'a été caractérisé.
Toxicology and risks
Aucune étude de toxicité réglementaire ni aucune donnée de sécurité chez l'être humain n'a été publiée. Le seul repère disponible est la classification de danger notifiée à l'Agence européenne des produits chimiques, qui range la substance en toxicité aiguë de catégorie 4 par voie orale, avec la mention « nocif en cas d'ingestion » : il s'agit d'un étiquetage de produit chimique de laboratoire, non d'une évaluation pharmacologique. Deux signaux issus de la recherche méritent d'être relevés. D'une part, chez la souris, une administration aiguë augmente des marqueurs de mort cellulaire dans le cortex, notamment l'activation de la kinase JNK, effet qui s'inverse après traitement répété, ce qui traduit un comportement de ligand protéen plutôt qu'un effet stable. D'autre part, l'action stimulante mise en évidence sur les îlots pancréatiques humains et murins, indépendante des récepteurs cannabinoïdes connus et du GPR55, montre que la sélectivité annoncée ne rend pas compte de tous les effets de la molécule. Rien dans la littérature ne permet d'estimer une marge de sécurité chez l'être humain, et aucune exposition humaine n'est justifiable.
Detection and analysis
Aucune méthode de dépistage de routine ne cible cette molécule. Elle n'est pas recherchée par les tests urinaires cannabinoïdes, qui visent les métabolites du THC, et ne partage avec eux ni structure ni métabolite. Elle ne figure pas davantage dans les panels consacrés aux cannabinoïdes de synthèse, conçus pour des agonistes des récepteurs CB1 et CB2 et non pour un agoniste inverse sélectif du CB2. En laboratoire, son dosage relèverait des techniques générales de chromatographie liquide couplée à la spectrométrie de masse ; un spectre obtenu en couplage chromatographie gazeuse et spectrométrie de masse est archivé dans les bases spectrales publiques, et le composé possède un identifiant unique de substance attribué par l'agence américaine du médicament, ce qui facilite son suivi documentaire. Aucune méthode médico-légale validée ne lui est consacrée.
