Pharmacology

JZL195 is a piperazine carbamate that covalently inactivates two serine hydrolases of the endocannabinoid system: FAAH, which degrades anandamide, and MAGL, which degrades 2-arachidonoylglycerol. Inhibition is potent, achieved at nanomolar concentrations on mouse brain membranes, and activity-based probe profiling indicates limited impact on the rest of the serine hydrolase proteome, with only modest inhibition of ABHD6. The molecule does not itself bind to CB1 or CB2: it acts indirectly, by raising the brain levels of both endocannabinoids strongly and durably. In mice, this dual blockade reproduces the cannabinoid tetrad, with analgesia, hypomotility and catalepsy, effects absent when FAAH or MAGL are inhibited separately, and it substitutes for THC in stimulus discrimination. Rimonabant, a CB1 antagonist, abolishes these responses, which confirms that they necessarily pass through the CB1 receptor. Human data are entirely lacking: neither pharmacokinetics, nor tolerance, nor safety of use. JZL195 remains a pharmacological tool for studying the interplay between anandamide and 2-arachidonoylglycerol, not a validated drug candidate.

Pharmacokinetics

Les seules données disponibles proviennent de la souris et du rat. Après administration systémique unique, JZL195 franchit la barrière hématoencéphalique et bloque la FAAH et la MAGL cérébrales pendant au moins une dizaine d'heures, avec une élévation des taux cérébraux d'anandamide et de 2-arachidonoylglycérol d'environ un ordre de grandeur, maintenue sur toute cette fenêtre. Aucune étude de pharmacocinétique humaine, de biodisponibilité orale ou de demi-vie plasmatique n'a été publiée.

Metabolism

Le motif carbamate constitue le groupe réactif : il carbamoyle la sérine catalytique de la FAAH et de la MAGL, ce qui rend l'inactivation covalente et de longue durée, la récupération de l'activité enzymatique dépendant alors de la resynthèse des protéines. Aucune identification de métabolites, aucune étude d'oxydation microsomale et aucune donnée d'excrétion n'ont été publiées, chez l'animal comme chez l'humain.

Toxicology and risks

Les données humaines manquent totalement. Chez la souris, une administration unique reproduit la tétrade cannabinoïde, avec analgésie, réduction de l'activité locomotrice et catalepsie, sans hypothermie ; ces effets sont abolis par le rimonabant, donc médiés par CB1. En discrimination de stimulus, JZL195 se substitue au THC, signal qui indique une psychoactivité de type cannabinoïde et un potentiel d'abus pertinent. Un traitement chronique chez le rat spontanément hypertendu n'a pas modifié le poids des organes, la glycémie, la température rectale ni les marqueurs plasmatiques de stress oxydant, mais ces observations chez le rongeur ne constituent pas une évaluation de sécurité transposable à l'humain. Aucun usage en dehors du laboratoire ne peut être considéré comme documenté.

Detection and analysis

Aucune méthode de dépistage de routine n'est documentée. JZL195 n'est ni un cannabinoïde ni un métabolite du THC : les immunoessais urinaires orientés cannabis et les criblages toxicologiques standards ne le détectent pas. Sa recherche supposerait une méthode ciblée par chromatographie liquide couplée à la spectrométrie de masse, avec un étalon de référence dédié. Aucun signalement au titre du système d'alerte précoce européen sur les nouvelles substances psychoactives n'a été identifié.
