Ses constantes de liaison figurent dans le travail de relation structure-activité publié par ce groupe en 2005 dans Bioorganic and Medicinal Chemistry, qui montrait qu'un groupe méthoxy en position 4 du noyau naphtoyle renforce l'affinité pour les deux récepteurs. Pharmacology
JWH-081 is a cannabinoid receptor agonist belonging to the naphthoylindole family, structurally alien to the cannabinoids of hemp. It binds to the CB1 receptor with nanomolar affinity, far greater than that of delta-9-tetrahydrocannabinol, and retains good affinity for the CB2 receptor. Where THC is only a partial agonist, naphthoylindoles of this type behave as high-efficacy agonists, which accounts for a more abrupt response and the absence of the effect ceiling observed with cannabis. Work in mice establishes that the impairment of recognition memory, the reduction of hippocampal long-term potentiation and the fall in phosphorylation of CaMKIV and CREB do indeed depend on the CB1 receptor, since they vanish in animals lacking that receptor. What the literature does not establish matters just as much: human data are lacking, no controlled study of dose, kinetics or safety exists, no therapeutic benefit is demonstrated, and any action on TRPV1, GPR55, PPARγ or 5-HT1A is undocumented for this molecule.

Origin (pharmaceutical research → illicit market)
No biological pathway exists for this molecule. JWH-081 is produced neither by hemp, nor by any plant, nor by the human body. It is built chemically from an indole core bearing a pentyl chain and a methoxynaphthoyl motif, without enzymatic involvement. Its presence in a plant product always results from impregnation after harvest.

Classified as a narcotic in France. JWH-081 is not named individually in any French text: it falls under the generic naphthoylindole clause, defined as derivatives of 3-(1-naphthoyl)indole, entered in annex IV of the decree of 22 February 1990 by the decree of 19 May 2015, which classified seven families of synthetic cannabinoids. Being a 1-pentyl-3-(4-methoxy-1-naphthoyl)indole, the molecule falls within this family definition. Possession, transfer, importation, exportation and use therefore fall under the criminal regime for narcotics. No food, cosmetic or wellness use is authorised.
European Union
No control measure specific to JWH-081 has been adopted at European Union level. The molecule was notified to the European early warning system in April 2010 and its status depends on national legislation, several member states covering it through generic definitions and others through lists of named substances. It does not appear in the schedules of the 1971 Convention on Psychotropic Substances, unlike JWH-018 and AM-2201, which were placed under international control in 2015. In the United States, it is listed by name in Schedule I under the 2012 federal law on synthetic drugs.

Pharmacokinetics
Les données humaines manquent : aucune étude contrôlée de pharmacocinétique n'a été publiée pour le JWH-081, et aucun paramètre validé de demi-vie, de biodisponibilité ou de volume de distribution n'est disponible. La seule voie d'exposition documentée est l'inhalation de mélanges végétaux imprégnés, dont la teneur varie fortement d'un lot à l'autre et parfois au sein d'un même sachet. La molécule est très lipophile, et plusieurs de ses métabolites hydroxylés restent pharmacologiquement actifs, si bien que la durée d'action ne se déduit pas de la disparition du composé parent dans le sang. Les concentrations sanguines, salivaires et capillaires rapportées proviennent de séries médico-légales et de cas isolés, jamais d'administrations contrôlées.
Metabolism
Métabolisme hépatique intense, caractérisé à partir d'urines de consommateurs et d'incubations in vitro. La voie majeure est la monohydroxylation, portant soit sur la chaîne N-pentyle, soit sur le noyau naphtalénique, soit sur le noyau indolique, accompagnée de métabolites carboxylés sur la chaîne alkyle. Les travaux de Hutter et de ses collègues n'ont pas mis en évidence de O-déméthylation du groupe méthoxy pour cette molécule, ce qui la distingue de certains analogues alkoxylés. Les métabolites hydroxylés sont ensuite conjugués à l'acide glucuronique par les UDP-glucuronosyltransférases avant excrétion urinaire. Comme pour d'autres naphtoylindoles, une partie de ces métabolites conserve une affinité pour le récepteur CB1, ce qui complique l'interprétation de la relation entre exposition et effet.
Toxicology and risks
Bannière de réduction des risques indispensable : il s'agit d'un cannabinoïde de synthèse, sans usage légal ni marge de sécurité établie chez l'être humain. Chez la souris, la dose la plus élevée testée provoque tremblements, réactions de traction, réduction de l'activité locomotrice et perte de performance au rotarod, avec des noyaux et des membranes nucléaires déformés dans la partie centrale du noyau accumbens, ce que les auteurs interprètent comme un signal de neurotoxicité. Une autre étude montre une altération de la reconnaissance d'objet, une baisse de la mémoire de travail spatiale et une suppression de la potentialisation à long terme hippocampique, effets absents chez les souris dépourvues de récepteur CB1. En clinique, les intoxications liées aux mélanges de type Spice contenant des naphtoylindoles associent tachycardie, agitation, comportements désorganisés, convulsions, et dans les séries de réanimation des défaillances respiratoires ayant conduit à l'intubation. Les produits saisis contiennent des mélanges de molécules à dosage inconnu et hétérogène, ce qui rend le risque de surdose élevé et imprévisible. Les données humaines contrôlées manquent totalement.
Detection and analysis
Le JWH-081 échappe aux immunoessais urinaires classiques du cannabis, qui ciblent les métabolites du THC : un dépistage négatif ne dit rien de son absence. La confirmation repose sur la chromatographie liquide couplée à la spectrométrie de masse en tandem ou à haute résolution. Dans l'urine, la molécule mère est rarement retrouvée, et les laboratoires ciblent les métabolites monohydroxylés, en particulier le dérivé N-(5-hydroxypentyl) qui dispose d'un code d'analyse dédié dans la nomenclature LOINC. Le cheveu offre une fenêtre rétrospective de plusieurs mois par analyse segmentaire, mais l'interprétation reste délicate : la fumée latérale et la simple manipulation des mélanges végétaux suffisent à contaminer la matrice, ce qui interdit de conclure à une consommation sur la seule présence du composé.
