JWH-133 is a molecule made entirely in the laboratory, absent from hemp and from any living organism. It was designed to bind almost exclusively to the CB2 receptor, the one found mainly in immune cells, and very little to the CB1 receptor of the brain. It is a research reagent: no study in humans exists and this substance is not found in any consumer product.

Pharmacology
JWH-133 is a classical cannabinoid from the series developed by John W. Huffman, described in 1999 as a 1-deoxy analogue of Δ8-THC bearing a 1,1-dimethylbutyl chain at position 3. Removal of the phenolic hydroxyl group collapses affinity for the CB1 receptor while preserving that for CB2: the published values are an inhibition constant of 3.4 nM at CB2 against 677 nM at CB1, that is, a selectivity ratio of about 200. The molecule behaves as a full agonist of CB2, coupled to Gi/o proteins, with inhibition of adenylate cyclase and modulation of the MAPK and NF-κB pathways; it is on this basis that it serves as a pharmacological tool for isolating the CB2 component in models of inflammation, pain and neuroinflammation, its receptor dependence having been confirmed by the disappearance of its effects in CB2-deficient mice. This selectivity, however, holds only within a narrow concentration window: at the micromolar scale, residual affinity for CB1 becomes relevant again. Human data are entirely lacking: no clinical trial, no human pharmacokinetic profile, no metabolism study and no regulatory toxicology study have been published, and the authors of the reference review explicitly call for prior safety work.

Origin (research tool)
No biosynthetic pathway: JWH-133 derives from no plant or animal metabolism. The compound comes exclusively from organic synthesis, by construction of the dibenzopyran skeleton of the classical cannabinoids with removal of the hydroxyl at position 1 and replacement of the pentyl chain by a 1,1-dimethylbutyl chain.

JWH-133 is listed by name on no ANSM list: the consolidated list of substances classified as narcotics cites many compounds of the Huffman series (JWH-018, JWH-073, JWH-250 and others) as well as the classical cannabinoids HU-210 and HU-243, but not JWH-133. The molecule does, however, fall under the generic clause of annex IV of the decree of 22 February 1990, introduced by the ANSM decision of 22 May 2024 and supplemented by that of 3 June 2024, which classifies as a narcotic any substance derived from the benzo[c]chromene core, hydrogenated or not on ring A, substituted at position 3 by an alkyl chain, at position 6 by one or two alkyl groups and at position 9 by an alkyl group. The 6,6,9-trimethyl-3-(2-methylpentan-2-yl)-6a,7,10,10a-tetrahydrobenzo[c]chromene skeleton matches this definition point by point and falls under none of the exclusions provided for (CBN, CBNA, THCV, THCVA, THCA). This is therefore a classification by generic clause and not a listing by name. It should be noted that the older clause covering tetrahydrocannabinols, their esters and their ethers does not apply here: the molecule is an analogue deoxygenated at position 1, neither an ester nor an ether of THC.
European Union
There is no harmonised control at European Union level: placing under control is a matter for national legislation, the European tier being limited to the early warning system and the risk assessment procedures provided for by Regulation (EU) 2017/2101 and Directive (EU) 2017/2103. JWH-133 has been the subject neither of a European risk assessment nor of a control measure at Union level, and it does not appear in the schedules of the 1961 and 1971 United Nations conventions. Several Member States apply generic definitions of synthetic cannabinoids whose scope differs from the one adopted in France, most often centred on the indole, indazole and cyclohexylphenol skeletons: the status of this molecule therefore varies appreciably from one country to another.

Cannabinoïde classique de type dibenzopyrane (noyau benzo[c]chromène), analogue 1-désoxy du Δ8-THC dont la chaîne pentyle est remplacée par une chaîne 1,1-diméthylbutyle en position 3. L'absence du groupe hydroxyle phénolique en position 1 est la modification structurale qui gouverne la sélectivité pour CB2. Une partie de la littérature désigne le composé comme un dérivé du Δ9-THC, mais la publication d'origine et le nom systématique retenu correspondent bien à la série Δ8 (6a,7,10,10a-tétrahydro).
Pharmacokinetics
Les seuls paramètres publiés proviennent de modèles animaux et de la revue de synthèse de 2021 : volume de distribution modéré, de l'ordre de 1 à 3 L/kg, et demi-vie courte, environ une heure. Les voies d'administration décrites en expérimentation sont intrapéritonéale, sous-cutanée, intraveineuse, intratumorale et orale, sur une plage de 0,05 à 20 mg/kg selon le modèle étudié. Aucune donnée de pharmacocinétique humaine, de biodisponibilité orale ou de passage de la barrière hémato-encéphalique n'a été publiée.
Metabolism
Aucune étude dédiée au métabolisme de JWH-133 chez l'humain n'a été publiée, ni sur hépatocytes ni sur microsomes hépatiques, et aucune enzyme de biotransformation n'a été identifiée pour cette molécule en particulier. Par analogie avec les cannabinoïdes partageant le même squelette dibenzopyrane, une oxydation microsomale de la chaîne latérale et du cycle terpénique suivie d'une conjugaison serait attendue, mais cette voie n'a pas été établie expérimentalement pour ce composé.
Toxicology and risks
Aucune étude de toxicologie réglementaire et aucun cas d'exposition humaine documenté ne sont disponibles pour JWH-133. Les travaux précliniques rapportent une tolérance apparente aux doses employées chez le rongeur, mais ces protocoles sont conçus pour mesurer une efficacité, pas une sécurité : la revue de référence conclut explicitement que des études de sécurité et de toxicité restent nécessaires avant tout passage chez l'humain. La sélectivité pour CB2 laisse attendre l'absence des effets psychotropes liés à CB1, ce que ne démontre aucune donnée humaine. Cette sélectivité n'est par ailleurs valable que dans une fenêtre de concentration étroite : à l'échelle micromolaire, l'affinité résiduelle pour CB1 redevient pharmacologiquement pertinente.
Detection and analysis
JWH-133 ne fait pas partie des panels de dépistage courants et n'est pratiquement pas rapporté dans les produits saisis, les agonistes CB2 sélectifs ne produisant pas l'effet psychotrope recherché sur le marché des cannabinoïdes de synthèse. Les immunoessais cannabinoïdes classiques, calibrés sur les métabolites du THC, ne le détectent pas. Son identification repose sur la chromatographie en phase gazeuse ou liquide couplée à la spectrométrie de masse, par comparaison avec un étalon de référence certifié. Aucun marqueur urinaire validé permettant d'attester une consommation n'a été publié à ce jour.
