La molécule appartient à la série des 1-[4-(pyridin-2-yl)benzyl]imidazolidine-2,4-diones décrite en 2011 dans le Journal of Medicinal Chemistry par Mario van der Stelt et ses coauteurs, alors dans la recherche pharmaceutique néerlandaise. Le code LEI, pour Leiden, a été attribué ensuite par le groupe de chimie médicinale de l'université de Leyde, et la caractérisation complète de LEI-101 a été publiée en 2016 dans le British Journal of Pharmacology par Partha Mukhopadhyay, Pál Pacher, Mario van der Stelt et leurs collaborateurs, avec des équipes du NIAAA aux États-Unis et de Rome. In plain terms

LEI-101 is a laboratory molecule with no link to hemp: it was designed to activate the CB2 receptor, the one found mainly on immune cells, while leaving aside the CB1 receptor of the brain. In the mouse, taken by mouth, it concentrates in the kidneys and the liver and causes no high. It has never been tried in human beings and is not a consumer product.

Pharmacology

LEI-101 is a partial agonist at the CB2 cannabinoid receptor, arising from a series of biaryl imidazolidine-2,4-diones designed in medicinal chemistry. At the human receptors, the reported affinity is pKi 7.5 for CB2 against less than 5 for CB1, with partial activation by both routes studied: 65% of the maximal effect in the [35S]GTPγS assay (pEC50 6.6) and 41% in β-arrestin recruitment (pEC50 7.0). The interest of the molecule lies above all in its distribution: administered orally in the mouse, it accumulates in the kidney and the liver and penetrates the brain very little, which explains the absence of any cannabimimetic sign in the tetrad test up to 60 mg/kg. In a model of nephrotoxicity induced by cisplatin, doses of 3 and 10 mg/kg reduced, in a dose-dependent manner, the impairment of renal function, the histological lesions, the oxidative and nitrative stress and the inflammation; the protection disappears in mice lacking the CB2 receptor, which clearly ties the effect to that target. All of this remains animal pharmacology: no clinical trial, no human pharmacokinetics and no human safety profile have been published. The molecule serves as a probe for studying the peripheral role of CB2, not as a treatment.

Origin (research tool)

No known biosynthetic pathway: LEI-101 comes from no plant or animal metabolism. It is a product of organic synthesis organised around a hydantoin (imidazolidine-2,4-dione) bearing a cyclopropyl, linked by a benzylic bridge to a fluorinated phenyl-pyridine biaryl, itself substituted by a thiomorpholine-1,1-dioxide motif. Its construction belongs to the chemistry of heterocycles and biaryl couplings, with no relation whatever to the cannabigerol pathway that gives rise to the hemp cannabinoids.

Hydantoïne (imidazolidine-2,4-dione) N-cyclopropylée d'un côté et N-benzylée de l'autre, le benzyle portant un biaryle phényle-pyridine fluoré substitué par une thiomorpholine-1,1-dioxyde ; la forme documentée est le chlorhydrate. Aucune parenté structurale avec les cannabinoïdes terpénophénoliques du chanvre ni avec les endocannabinoïdes de type amide d'acide gras.

Pharmacokinetics

Les données disponibles proviennent uniquement de la souris. L'absorption orale est bonne et les concentrations plasmatiques culminent environ deux heures après la prise, par voie orale comme par voie intrapéritonéale. La distribution est franchement périphérique : les concentrations les plus élevées se retrouvent dans le rein et le foie, les plus faibles dans le cerveau, ce qui fonde la notion de restriction périphérique et explique l'absence d'effets centraux. La fraction plasmatique libre rapportée est de 0,44. Aucune donnée humaine n'existe : ni demi-vie, ni biodisponibilité absolue, ni comportement en administration répétée.

Metabolism

Le devenir métabolique de LEI-101 n'a pas été publié : les voies de biotransformation, les enzymes impliquées et les métabolites restent non caractérisés, chez l'animal comme chez l'humain. Le profilage par sondes d'activité conduit sur les sérine hydrolases de l'endocannabinoïdome n'a montré aucune inhibition de la FAAH, de la MAGL, de la NAPE-PLD ou de la DAGL, mais ce résultat porte sur la sélectivité de la molécule et non sur son élimination.

Toxicology and risks

Aucune évaluation toxicologique formelle n'a été publiée et la molécule n'a jamais été administrée à l'humain. Le seul élément de tolérance disponible relève de la pharmacologie de sécurité comportementale chez la souris : au test de la tétrade, une administration orale allant jusqu'à 60 mg/kg n'a produit ni analgésie, ni hypothermie, ni catalepsie, ni baisse de l'activité locomotrice, autrement dit aucun effet central de type cannabinoïde. Les expériences d'efficacité rénale ne rapportent pas d'effet indésirable aux doses actives. Faute d'étude de toxicité à doses répétées, de génotoxicité, de sécurité cardiaque et de toute donnée humaine, aucun profil de risque ne peut être établi.

Detection and analysis

Aucune méthode de dépistage de routine ne cible cette molécule : elle n'a pas d'usage récréatif documenté, n'a jamais été signalée sur le marché des nouvelles substances psychoactives et ne figure dans aucun panel toxicologique urinaire ou salivaire. Les mesures publiées reposent sur des dosages bioanalytiques dédiés par chromatographie liquide couplée à la spectrométrie de masse, réalisés sur le plasma et les tissus de souris. Aucune littérature de médecine légale ne lui est consacrée.
