Le lysophosphatidylinositol est décrit comme constituant lipidique dès les années 1960, puis rapporté au milieu des années 1980 par Metz comme stimulant de la sécrétion d'insuline par les îlots pancréatiques. Son statut de médiateur s'établit dans les années 1990 avec les travaux de Marco Falasca et Daniela Corda, qui montrent son accumulation et son action mitogène dans des cellules thyroïdiennes transformées par l'oncogène Ras, puis sa libération dans le milieu par des fibroblastes transformés. Le tournant survient en 2007 : Saori Oka, Takayuki Sugiura et leurs collègues, au Japon, identifient le LPI comme agoniste du récepteur orphelin GPR55. La même équipe désigne en 2009 le 2-arachidonoyl-LPI comme l'espèce la plus puissante et le ligand naturel le plus probable. La structure du complexe GPR55/LPI est résolue par cryomicroscopie électronique en 2025 par Tobias Claff, Dietmar Weichert et leurs collègues.

LPI is a normal constituent of mammalian cell membranes: it arises from the partial hydrolysis of phosphatidylinositols in practically every tissue and circulates in human plasma at concentrations of the order of a few micromoles per litre. Its levels vary with the pathological context: they are elevated in Ras-transformed cells, in the plasma of patients with ovarian cancer, in obese individuals and in acute coronary syndrome. There is no plant source of interest for food or cosmetic use; the preparations used in the laboratory are obtained by enzymatic hydrolysis of soya phosphatidylinositols using a phospholipase A2, which yields a mixture of species dominated by the palmitoylated 16:0 form, the one described in this entry.

LPI is a fragment of membrane fat that the cells of the human body make by cutting up a phospholipid; it circulates in the blood and serves as a signal between cells. It binds to a receptor called GPR55, often presented as a distant cousin of the cannabis receptors, but it does not act on the classic cannabinoid receptors. It is not found in hemp, goes into no consumer product, and everything known about it comes from the laboratory.

Pharmacology

LPI occupies a singular place in the literature on the endocannabinoid system: it is the reference endogenous ligand of GPR55, a receptor long presented as a third cannabinoid receptor, even though the molecule itself is in no way a cannabinoid and activates neither CB1 nor CB2. Its identification dates back to 2007, when Oka and colleagues showed that LPI, alone among the lysolipids tested, triggers ERK1/2 phosphorylation, calcium transients and GTPγS binding in cells expressing GPR55. The structure of the complex, obtained by cryo-electron microscopy in 2025, specifies the anchoring of the phosphate to arginine 253, the bonds of the inositol head and the exit of the fatty chain towards the membrane, with selective coupling to Gα13 and an EC50 of 35.5 nM for the 16:0 species described here; the 2-arachidonoyl species remain the most potent and are regarded as the most probable natural ligand. Downstream, RhoA and ROCK reorganise the cytoskeleton, phospholipase C mobilises calcium, and ERK relays towards transcription. Not everything is attributable to GPR55: in enteroendocrine L cells, the GLP-1 secretion induced by LPI depends on the TRPV2 channel and is not reproduced by selective agonists of the receptor. The relationship with plant cannabinoids is above all inhibitory, with cannabidiol as well as THCV and CBDV blocking LPI-triggered signalling. Human data are lacking: no controlled administration, no clinical trial, only measurements of endogenous concentrations and associations with obesity, ovarian cancer or acute coronary syndrome. No therapeutic benefit is established.

Biosynthetic pathway (in vivo)

LPI arises from the partial hydrolysis of membrane phosphatidylinositols rather than from a dedicated synthetic pathway. A calcium-dependent cytosolic phospholipase A2 removes the fatty chain at position 2 and releases a 1-acyl-LPI: this is the case for the 16:0 species described here, which retains its palmitate. A phospholipase A1, DDHD1, acts in the opposite sense, removing the chain at position 1 and releasing a 2-acyl-LPI, in particular the 2-arachidonoyl-LPI derived from 1-stearoyl-2-arachidonoyl phosphatidylinositol: it is this species, and not the 16:0 form, that is most often cited as the physiological agonist of GPR55. Production is tied to the state of the cell rather than constitutive: it increases markedly in Ras-transformed cells, which release it into the extracellular medium.

Lysophospholipide de la famille des glycérophosphoinositols : un squelette glycérol portant une seule chaîne grasse estérifiée, l'autre position hydroxyle étant libre, et un groupement phosphate reliant le glycérol à un cycle myo-inositol. La fiche correspond à l'espèce 16:0, dont la chaîne est un palmitate ; l'espèce la plus citée pour son activité est le 2-arachidonoyl-LPI, qui porte à la place un résidu arachidonoyle à vingt carbones et quatre insaturations. Ce n'est pas un cannabinoïde au sens chimique : la molécule ne possède ni noyau terpénophénolique, ni fonction amide de type N-acyléthanolamine, ni ester de glycérol comparable au 2-AG.

Pharmacokinetics

Aucune étude de pharmacocinétique humaine n'a été publiée : le LPI n'a jamais été administré à l'être humain, et rien n'est documenté sur une éventuelle absorption orale, qui se heurterait de toute façon aux lipases et aux phospholipases digestives. Ce que la littérature mesure, ce sont des concentrations endogènes : les travaux sur les cellules entéro-endocrines situent le LPI circulant autour de trois micromoles par litre chez l'adulte, ordre de grandeur cohérent avec les concentrations actives in vitro. Moreno-Navarrete et ses collègues (2012) rapportent des taux plasmatiques plus élevés chez des patientes obèses, corrélés au poids et au pourcentage de masse grasse, avec une expression accrue de GPR55 dans le tissu adipeux. Il s'agit d'un médiateur local produit à la demande et non stocké, dont la persistance dans le milieu extracellulaire est limitée par des hydrolases et par la réacylation en phospholipide.

Metabolism

Le LPI est à la fois un produit de dégradation des phosphatidylinositols et un substrat pour plusieurs voies. Une lysophospholipase A libère l'acide gras et donne le glycérophosphoinositol ; une lysophospholipase C conduit à des acylglycérols ; une lysophospholipase D, l'autotaxine (ENPP2), coupe la tête inositol et transforme le LPI en acide lysophosphatidique, médiateur qui possède ses propres récepteurs, ce qui complique l'attribution des effets observés. La voie de recyclage est la réacylation en phosphatidylinositol par des acyltransférases : la MBOAT7 et l'AGPAT8 prennent en charge le 1-acyl-LPI, seule l'AGPAT8 réacylant le 2-acyl-LPI. Des travaux récents décrivent en outre un catabolisme hépatique du LPI relié à la transduction du signal insulinique. Aucune donnée humaine d'élimination ni de bilan de masse n'existe, puisque la molécule n'a jamais été administrée.

Toxicology and risks

Aucune donnée de toxicologie humaine, aucun cas d'intoxication, aucun potentiel d'abus : le LPI n'a pas d'effet psychotrope connu, n'agit pas sur CB1 et n'est pas une substance de consommation. Les signaux disponibles sont des associations pathologiques et des expériences animales, et ils vont dans les deux sens. Les concentrations de LPI sont élevées dans les cellules transformées par Ras et dans le plasma de patientes atteintes de cancer de l'ovaire, et l'axe LPI/GPR55 favorise la prolifération et la migration de plusieurs lignées tumorales ; chez le rongeur, l'administration de LPI aggrave les lésions d'ischémie-reperfusion myocardique par une voie GPR55/ROCK. À l'inverse, des effets anti-inflammatoires ont été décrits sur microglie en culture, et une amélioration de la sécrétion d'insuline et de la tolérance au glucose sur modèles animaux. Le sens de l'effet dépend du tissu et du contexte, ce qui interdit toute conclusion générale et tout usage extrapolé chez l'être humain.

Detection and analysis

Quantification par chromatographie liquide couplée à la spectrométrie de masse, généralement en tandem et avec étalons internes, dans les panels lipidomiques qui dosent conjointement les lysophospholipides plasmatiques : LPI, acide lysophosphatidique, lysophosphatidylcholine et lysophosphatidylsérine. Les espèces se distinguent par la longueur et l'insaturation de la chaîne grasse, 16:0, 18:0 ou 20:4, ce qui impose une séparation chromatographique fine avant détection. La mesure reste délicate : le LPI continue d'être produit et dégradé après le prélèvement, si bien que le traitement des échantillons doit être rapide et que les valeurs absolues se comparent mal d'une étude à l'autre. L'usage est strictement scientifique ou clinique exploratoire ; le LPI n'apparaît dans aucun test de dépistage toxicologique ni dans aucune recherche médico-légale, puisqu'il s'agit d'un lipide endogène non contrôlé.
