- La molécule n'a pas d'inventeur académique : elle est apparue directement sur le marché des mélanges à fumer. L'administration antidrogue des États-Unis situe sa première identification dans des saisies en novembre 2012, et le laboratoire criminalistique de l'armée américaine a rapporté ses premiers cas positifs en octobre 2014. Sur le plan scientifique, Shevyrin et ses collaborateurs ont décrit en novembre 2014 la série des indole-3-carboxylates apparus dans les mélanges à fumer, puis Kondrasenko, Goncharov, Dugaev et Rubaylo ont publié en décembre 2015 un rapport dédié au CBL-2201, réunissant les données de résonance magnétique nucléaire, d'infrarouge et de chromatographie gazeuse nécessaires à son identification. La nomenclature systématique retenue par l'observatoire européen la désigne également par le code NA-5F-PIC.

The powder is distributed unevenly across the substrate, so that two doses taken from the same sachet may deliver very different quantities.

In plain terms
NM-2201, also sold under the code CBL-2201, is a synthetic cannabinoid manufactured in clandestine laboratories. It has no connection with hemp or with cannabidiol products: it is sprayed onto smoking herbs or added to vaping liquids, and it acts on the same receptors as THC, but far more strongly. Serious hospitalisations have been associated with it, and it is classified as a narcotic in France under its own name.

Pharmacology
NM-2201 is the naphthalen-1-yl ester of 1-(5-fluoropentyl)indole-3-carboxylic acid, a structural twin of 5F-PB-22 from which it differs only in the replacement of the quinoline ring by a naphthalene. Hess and colleagues (Forensic Toxicology, 2016) rank it among the highest-affinity constituents of "spice": a Ki of 0.332 nM at the human CB1 receptor and 0.732 nM at CB2, with no selectivity between the two subtypes, and, in a cAMP accumulation assay, an efficacy greater than that of the reference full agonist CP55,940. Δ9-THC, measured in the same system, is only a partial agonist and of markedly lower affinity: this gap in potency and efficacy is the heart of the risk. The same study finds no activity at the orphan receptors GPR18 and GPR55, and nothing has been published on TRPV1, PPARγ or 5-HT1A. In mice, the molecule reproduces the four effects of the cannabinoid tetrad, induces a conditioned place preference at low dose and then an aversion at higher dose, and a CB1 antagonist precipitates a withdrawal syndrome. Beyond that, human data are lacking: no clinical study, no established dose-effect relationship, no pharmacokinetics measured in human beings. The available knowledge rests on in vitro work, on animal models and on forensic observations.

Origin (pharmaceutical research → illicit market)
No known biosynthetic pathway: NM-2201 is produced by no plant and no living organism. It arises solely from clandestine organic synthesis, through esterification of an indole-3-carboxylic acid bearing a 5-fluoropentyl chain on the indole nitrogen with naphthalen-1-ol. The description here is deliberately confined to the chemical class.
Legal framework
France
NM-2201 is classified as a narcotic in France, and it is so by name. The decree of 31 March 2017 (NOR AFSP1710288A) added to annex IV of the decree of 22 February 1990 a structural family entitled "Indole-3-carboxylate derivatives", covering indoles bearing on the nitrogen an alkyl, haloalkyl, halobenzyl, alkenyl, cycloalkylmethyl, cycloalkylethyl or related substituent, and on the oxygen of the carboxylate bridge an 8-quinolinyl or 1-naphthalenyl group. The text then expressly cites "NM-2201 or CBL-2201", alongside PB-22, BB-22, 5F-PB-22, FUB-PB-22 and FDU-PB-22. This is therefore not a capture by the generic clause covering tetrahydrocannabinols and their esters: the substance does indeed appear under its own name. Production, importation, possession, transfer and use fall under the criminal regime for narcotics, and no product legally sold in France may contain it.
European Union
No European Union act names NM-2201: it does not appear in the annex to Framework Decision 2004/757/JHA, and its control is a matter for national law. Germany lists it in annex II of its narcotics law, Sweden classified it following the November 2014 proposal of its public health agency, and France names it in its own decree. The United Kingdom covers it through the generic class B definition in its misuse of drugs legislation. Internationally, the molecule appears neither in the schedules of the 1961 Single Convention nor in those of the 1971 Convention on Psychotropic Substances: there is no worldwide control measure. Outside Europe, the United States placed it in Schedule I on a temporary basis on 10 July 2018, then permanently on 10 June 2021 under code 7221; Canada lists it in Schedule II of its Controlled Drugs and Substances Act; Japan had already added it to its list before 2016; and China has applied since 2021 a class-wide ban covering this structural family.

Ester naphtalén-1-ylique de l'acide 1-(5-fluoropentyl)indole-3-carboxylique, sous-famille des 3-carboxylates d'indole parmi les agonistes de synthèse des récepteurs cannabinoïdes. Il ne diffère du 5F-PB-22 que par le remplacement du noyau quinoléine par un noyau naphtalène, et de l'AM-2201 que par le remplacement du pont cétone par un pont ester. Le code commercial CBL-2201 désigne la même molécule et n'a aucun rapport avec le cannabicyclol, dont l'abréviation usuelle est CBL.
Pharmacokinetics
Molécule très lipophile : les prédictions in silico publiées par Hess et ses collaborateurs la classent parmi les composés capables de franchir la barrière hémato-encéphalique. Le pont ester constitue le point faible du squelette. Sur microsomes hépatiques humains, la demi-vie mesurée n'est que de huit minutes, l'hydrolyse étant attribuée aux carboxylestérases hépatiques. La molécule mère n'a pas été retrouvée dans les urines de sujets dont l'exposition était pourtant confirmée, ce qui oblige à rechercher les métabolites plutôt que le composé parent. Aucune étude de pharmacocinétique humaine n'a été conduite : ni demi-vie plasmatique, ni biodisponibilité, ni volume de distribution ne sont établis. Les usagers rapportent sur des forums une durée d'effet plus courte que celle du JWH-018, mention qui n'a évidemment aucune valeur de mesure.
Metabolism
Diao et ses collaborateurs ont caractérisé le métabolisme humain sur hépatocytes, puis vérifié leurs résultats sur deux urines authentiques de sujets exposés. Treize métabolites ont été identifiés. La voie dominante est l'hydrolyse du pont ester, qui libère d'un côté le naphtalén-1-ol, excrété sous forme de glucuronide, et de l'autre l'acide 1-(5-fluoropentyl)indole-3-carboxylique, désigné 5F-PI-COOH. S'y ajoutent des hydroxylations sur la chaîne fluoropentyle et sur le noyau indole, une défluoration oxydative et des glucuronoconjugaisons. Dans les urines, ce même acide est le métabolite prédominant après hydrolyse enzymatique des conjugués, et les auteurs le retiennent comme marqueur urinaire de référence. Ce marqueur présente une limite importante : le 5F-PB-22 produit exactement le même métabolite, si bien que sa détection isolée ne permet pas de dire laquelle des deux substances a été consommée.
Toxicology and risks
Aucune étude toxicologique réglementaire n'existe, et aucun décès avec exposition analytiquement confirmée au seul NM-2201 n'a été retrouvé dans la littérature consultée. Les signaux disponibles restent néanmoins sérieux. L'administration antidrogue des États-Unis relie directement à cette substance des passages aux urgences associant sudation profuse, tachycardie, hypertension artérielle, convulsions, agitation, violence, nausées et troubles de la mémoire. Elle lui rattache également un épisode collectif survenu à Ocala, en Floride, en décembre 2015 : vingt-cinq à trente personnes ont été conduites à l'hôpital après des scènes de violence et des convulsions. Un cas britannique publié en 2017 décrit un homme de vingt-cinq ans sans antécédent, hospitalisé après avoir fumé un produit dont l'analyse a confirmé qu'il contenait du NM-2201 : aphasie, hémiparésie gauche, hypertonie, puis fièvre, sudation profuse, tachycardie, poussées hypertensives et probable crise convulsive, avec des lésions cérébrales multifocales évoquant une encéphalomyélite aiguë disséminée, quatorze jours de réanimation et une trachéotomie avant une récupération lente. Chez la souris, une étude a établi une dose létale médiane, retrouvé les quatre effets de la tétrade cannabinoïde et mis en évidence un syndrome de sevrage précipité par un antagoniste de CB1. Le risque principal tient à l'impossibilité de connaître la dose reçue : la poudre est répartie de façon inégale sur le support végétal et rien n'indique la composition réelle du produit. Les cas rapportés associent presque toujours plusieurs substances.
Detection and analysis
Les immunodosages cannabis usuels ne détectent pas cette molécule. Son identification dans les saisies est délicate : Kondrasenko et ses collaborateurs ont montré qu'une recherche automatique en bibliothèque de spectres de masse ne permet pas de la distinguer de façon fiable du dérivé quinoléin-8-ylique correspondant, c'est-à-dire du 5F-PB-22, et qu'une confirmation par résonance magnétique nucléaire à une et deux dimensions reste nécessaire. D'autres approches ont été validées pour discriminer ces analogues proches, notamment la chromatographie gazeuse couplée à une ionisation électronique froide, et la spectroscopie Raman associée à une analyse chimiométrique, cette dernière regroupant toutefois le NM-2201 et le FDU-PB-22 du fait de leur ressemblance structurale. En matrice biologique, la recherche repose sur la chromatographie liquide couplée à la spectrométrie de masse haute résolution et cible l'acide 5F-PI-COOH, la molécule mère étant absente des urines.
