It was first demonstrated in the bovine striatum, then found in central and peripheral nervous tissue of the rat, where it results from the assembly of dopamine and oleic acid in catecholaminergic neurons. The samples used in research are of synthetic origin and sold as laboratory reagents, never as ingredients of consumer products.

In plain terms

OLDA is a molecule the body makes itself, by joining dopamine to a very common fatty acid, oleic acid, the one in olive oil. It was spotted in 2003 in the mammalian brain. It does not come from hemp and is not an ingredient of any CBD product: it is a research substance, known chiefly because it activates the sensor for the burning sensation, the same one triggered by chilli.

Pharmacology

OLDA belongs to the N-acyl dopamines, a family of endogenous lipids close to NADA. Its profile is vanilloid first rather than cannabinoid: it activates the TRPV1 channel with nanomolar potency, greater than that of capsaicin in several animal models, whereas its activity at the CB1 receptor is markedly weaker and nothing is documented at CB2. Work published since 2003 also credits it with activation of the GPR119 receptor, a modest blockade of dopamine reuptake and a modulation of midbrain dopaminergic neurons. The effects observed, thermal hyperalgesia after spinal administration, protection of the heart against ischaemia-reperfusion, induction of interleukin 10 in sepsis models, disappear in animals lacking TRPV1, which clearly designates that channel as the principal relay. All these data come from rodents or cell cultures: human data are lacking, no clinical trial has been conducted and no therapeutic benefit is established in humans.

Biosynthetic pathway (in vivo)

Formation by condensation of the carboxyl group of oleic acid with the amine function of dopamine, within catecholaminergic neurons. For the arachidonic congener NADA, direct conjugation of the fatty acid and dopamine is the accepted route, the contribution of an N-acyl tyrosine precursor transformed by tyrosine hydroxylase remaining debated. No synthetic enzyme dedicated to OLDA has been characterised with certainty.

N-acyl-dopamine, amide gras rangé par LIPID MAPS dans les N-acyl-amines (FA0802) : une chaîne oléoyle en C18 mono-insaturée de configuration Z, reliée par une liaison amide au groupe amine de la dopamine, dont le noyau catéchol porte le motif vanilloïde. Ce n'est pas un cannabinoïde terpénophénolique, et la molécule ne partage aucun squelette avec le CBD ou le THC.

Pharmacokinetics

Les paramètres humains sont inconnus. Chez le rongeur, l'OLDA apportée de façon exogène est captée par le tissu nerveux central et périphérique, et reste détectable jusqu'à vingt-quatre heures après administration sans trace d'hydrolyse ni d'auto-oxydation, ce qui la rend plus stable in vitro que la dopamine elle-même. Sa forte lipophilie, avec un logP calculé élevé et deux règles de Lipinski enfreintes, explique son association aux membranes neuronales ; elle est par ailleurs reconnue par le transporteur membranaire de l'anandamide. Aucune donnée de biodisponibilité orale, de distribution tissulaire chiffrée ou de demi-vie plasmatique n'est publiée.

Metabolism

La voie principale documentée est la O-méthylation du noyau catéchol par la catéchol-O-méthyltransférase, démontrée in vitro, ex vivo et in vivo chez le rat, et supprimée de façon dose-dépendante par le tolcapone, un inhibiteur de cette enzyme. Le métabolite O-méthylé reste minoritaire, de l'ordre de quelques pour cent de la quantité administrée. Contrairement au NADA, l'OLDA n'interagit pas avec l'amide hydrolase des acides gras (FAAH), ce qui écarte en pratique l'hydrolyse de la liaison amide comme voie majeure. L'oxydation du catéchol en quinone n'a pas été observée dans le tissu nerveux de mammifère.

Toxicology and risks

Aucune donnée humaine, aucun essai clinique, aucun cas d'exposition rapporté. Les études animales n'ont pas signalé de toxicité aux quantités employées, mais aucune n'a été conçue comme une étude de sécurité et aucun protocole réglementaire de toxicologie n'a été publié. Le signal biologique le plus net est pro-nociceptif : l'administration spinale raccourcit la latence de retrait à la chaleur chez le rat, et le TNF alpha sensibilise les récepteurs TRPV1 de la moelle à des concentrations d'OLDA autrement inactives, ce qui rattache la molécule aux mécanismes de la douleur inflammatoire plutôt qu'à un effet apaisant. Aucun usage humain, alimentaire ou cosmétique n'est autorisé.

Detection and analysis

L'identification initiale dans le tissu cérébral a reposé sur la spectrométrie de masse à temps de vol couplée à un quadripôle, appliquée à des extraits de striatum. Le dosage courant se fait par chromatographie liquide couplée à la spectrométrie de masse en tandem, en ionisation négative et en suivi de réactions multiples, avec des transitions distinctes pour le composé parent et pour son métabolite O-méthylé. Aucun dépistage toxicologique de routine ne recherche l'OLDA, et sa présence physiologique dans les tissus rendrait de toute façon un résultat positif ininterprétable en contexte médico-légal.
