- Le composé a été isolé en 1994 par Yoshinori Asakawa et ses collègues de l'université de Tokushima Bunri, à partir de l'hépatique japonaise Radula perrottetii, puis retrouvé chez Radula marginata, espèce endémique d'Aotearoa Nouvelle-Zélande. Il est resté une curiosité de phytochimie des bryophytes pendant près d'un quart de siècle. Sa pharmacologie n'a été établie qu'en 2018, quand Andrea Chicca, Michael Schafroth et leurs collègues des universités de Berne et de l'École polytechnique fédérale de Zurich en ont réalisé la synthèse totale et démontré l'activité cannabinoïde chez la souris, dans Science Advances. Le travail était motivé par la vente en ligne de préparations de Radula présentées comme des substituts légaux du cannabis, commerce alors dépourvu de toute donnée pharmacologique. Une caractérisation phytochimique complète de Radula marginata a suivi en 2025 dans New Phytologist.

In plain terms
Perrottetinene is a natural cousin of THC, but it does not come from hemp: it is made by liverworts, small plants related to mosses. It acts on the same receptors as THC, binding to them about ten times less strongly, and in mice it produces comparable but attenuated effects. No study has been carried out in humans, and in France it has fallen since 2024 under a generic clause of the narcotics list.

Pharmacology
Perrottetinene is the only psychoactive cannabinoid known outside hemp. Produced by liverworts of the genus Radula, it reproduces the benzo[c]chromene skeleton of tetrahydrocannabinol but replaces the pentyl chain with a bibenzyl chain, and adopts the cis configuration where hemp tetrahydrocannabinol is trans. Isolated in 1994, it remained without established pharmacology until the total synthesis published in 2018, which showed that both diastereoisomers cross the blood-brain barrier and produce hypothermia, catalepsy, hypolocomotion and analgesia in mice, effects abolished by CB1 blockade. Affinity remains modest: 481 nM at CB1 for the natural cis product, against 22 nM for tetrahydrocannabinol in the same assay, and efficacy plateaus at 60 or 80 % of that of a full agonist. The brain-to-plasma concentration ratio, on the other hand, is more favourable for the natural product than for tetrahydrocannabinol, which explains why a net effect is observed despite the lower affinity. The most original feature lies in the action on prostaglandins: both diastereoisomers lower basal brain levels of prostaglandins D2 and E2 through a CB1-dependent mechanism, whereas tetrahydrocannabinol does not, which prompted speculation about an anti-inflammatory profile with fewer adverse effects. None of this has been verified in humans: there is no clinical study, no data on dose, tolerance or safety, and the Radula preparations sold online do not declare their content.

Biosynthetic pathway
The plant pathway has not been elucidated in detail, but its architecture parallels that of hemp rather than being identical to it. The skeleton is not terpenophenolic in the strict sense: the aromatic ring bears a bibenzyl chain at position 3, that is, a 2-phenylethyl group, where tetrahydrocannabinol bears a linear pentyl chain. This unit arises from bibenzyl metabolism, which is highly developed in liverworts, and not from the fatty-chain polyketide pathway that supplies the olivetolic acid of hemp. The terpene portion and the closure of the pyran ring, by contrast, reproduce the tetrahydrocannabinol motif. The natural product is the cis diastereoisomer, the configuration opposite to that of hemp tetrahydrocannabinol, which is trans.
Legal framework
France
No listing by name, but a generic clause has applied since 2024. The ANSM decision of 22 May 2024 added to the narcotics list any substance derived from the benzo[c]chromene ring, hydrogenated or not on ring A, bearing at position 1 a hydroxyl function, at position 3 an alkyl chain optionally substituted by one or more cyclic groups, at position 6 one or two alkyl groups and at position 9 an alkyl group. Perrottetinene meets this definition point by point, its 2-phenylethyl chain being precisely an alkyl chain bearing a cyclic substituent. It falls within none of the exclusions provided by the text, which cover only cannabinol, cannabinolic acid, tetrahydrocannabivarin, tetrahydrocannabivarinic acid and tetrahydrocannabinolic acid. As with any classification by generic clause, this reading remains more fragile before a court than a listing by name.
European Union
No control measure at European Union level targets perrottetinene by name, and the molecule has been the subject of neither a risk assessment nor a Council implementing decision. It appears in no schedule of the 1961 Convention on narcotic drugs or the 1971 Convention on psychotropic substances: the international listings relating to tetrahydrocannabinol cover precisely defined pentyl-chain isomers, to which the bibenzyl chain does not correspond. Radula preparations therefore circulate in several member states as plant products, a status that depends on national generic clauses, which vary widely from one country to another. The 2018 work already noted that this trade was taking place without any safety data.

Cannabinoïde bibenzylique. Le squelette est celui du tétrahydrocannabinol, un noyau benzo[c]chromène partiellement hydrogéné portant une fonction phénol en position 1 et deux méthyles géminés en position 6, mais la chaîne latérale en position 3 est un groupe 2-phényléthyle au lieu de la chaîne pentyle. Le produit naturel est le diastéréoisomère cis, de configuration 6aS,10aR, alors que le tétrahydrocannabinol du chanvre est trans, de configuration 6aR,10aR. La molécule appartient donc aux cannabinoïdes au sens pharmacologique, mais non aux phytocannabinoïdes du chanvre au sens botanique.
Pharmacokinetics
Aucune donnée humaine. Chez la souris, la caractérisation de 2018 rapporte un rapport des concentrations cerveau sur plasma de 0,91 pour le produit naturel cis, contre 0,51 pour le diastéréoisomère trans et 0,51 pour le tétrahydrocannabinol trans, ce qui indique une pénétration cérébrale au moins aussi bonne que celle du tétrahydrocannabinol et probablement meilleure pour la forme naturelle. Les effets comportementaux ont été obtenus par voie intrapéritonéale à 50 mg/kg pour le diastéréoisomère cis et 40 mg/kg pour le trans, à comparer aux 10 mg/kg de tétrahydrocannabinol employés comme référence : il faut donc quatre à cinq fois plus de matière pour un effet du même ordre, ce qui reflète l'affinité plus faible. Aucune biodisponibilité orale, aucune demi-vie plasmatique et aucun volume de distribution ne sont établis, chez l'animal comme chez l'être humain.
Metabolism
Le devenir métabolique n'a pas été caractérisé. Aucun métabolite n'a été identifié, ni chez l'animal ni sur microsomes hépatiques humains, et les enzymes en cause ne sont pas connues. Par analogie de structure, les positions oxydables attendues seraient l'hydroxylation allylique du méthyle en position 9 et l'oxydation de la chaîne bibenzylique, mais aucune publication ne le documente et il ne s'agit donc que d'une extrapolation. Il n'est pas non plus établi si des métabolites actifs se forment, question centrale pour le tétrahydrocannabinol dont le dérivé 11-hydroxylé est plus actif que la molécule mère.
Toxicology and risks
Aucune étude toxicologique réglementaire, aucun essai clinique et aucun cas d'intoxication publié. Les seules données proviennent de l'expérimentation animale de 2018, où des doses uniques élevées par voie intrapéritonéale ont produit la tétrade cannabinoïde classique, hypothermie, catalepsie, hypolocomotion et analgésie, sans que la mortalité, la tolérance ou les effets d'une exposition répétée soient étudiés. Le contexte d'exposition réel est la vente en ligne de préparations de Radula présentées comme substituts légaux du cannabis : la teneur y est inconnue, très variable selon l'espèce et le lot, et aucune information de sécurité n'accompagne ces produits. Aucune dose humaine n'est caractérisée, et l'extrapolation à partir des doses murines n'aurait aucune valeur.
Detection and analysis
Aucune recherche de routine ne vise le perrottétinène. Il échappe aux immunoessais destinés au cannabis, qui ciblent le métabolite acide du tétrahydrocannabinol, et il n'entre dans aucun panel toxicologique courant. Son identification relève de la chromatographie en phase gazeuse ou liquide couplée à la spectrométrie de masse, avec une difficulté propre : la séparation des diastéréoisomères cis et trans, qui partagent la même masse et le même profil de fragmentation et exigent une séparation chromatographique adaptée. La caractérisation phytochimique de Radula marginata publiée en 2025 dans New Phytologist fournit les profils analytiques de référence pour la matière végétale et les bibenzyles qui l'accompagnent.
