- Décrit en 2007 par Horswill et ses collaborateurs dans le British Journal of Pharmacology, comme le premier antagoniste allostérique du récepteur CB1 pour lequel une activité in vivo a été démontrée. In plain terms
PSNCBAM-1 is a laboratory molecule with no connection whatsoever to hemp: it is found in no plant and goes into no consumer product. Researchers use it to restrain the brain's CB1 receptor without acting at the site targeted by cannabinoids themselves, in order to better understand how that receptor works. It has never been administered to human beings.

Pharmacology
PSNCBAM-1 is a negative allosteric modulator of the CB1 receptor: it binds at a site distinct from the orthosteric pocket, the one also shared by Org 27569, and lowers the efficacy of agonists instead of occupying their binding site. Horswill and co-workers showed in 2007 that it reduces, non-competitively as verified by Schild analysis, the responses to CP 55,940, WIN 55,212-2, anandamide and 2-AG, with no effect at all on the CB2 receptor. Its profile remains paradoxical: it increases [3H]CP 55,940 binding, measured at around 159 per cent of control, while decreasing the signalling that follows from it, and its action depends on the probe used, markedly more pronounced against CP 55,940 than against WIN 55,212-2. In cerebellar electrophysiology, it abolishes the reduction in the frequency of miniature inhibitory currents induced by CP 55,940 without altering that induced by WIN 55,212-2. In vivo, it lowers food intake and weight in the rat, and reduces ethanol self-administration in the mouse through a non-specific hypophagic effect, without altering conditioned place preference. Human data are entirely lacking: no clinical study, no human pharmacokinetics, no established safety profile.

No biological pathway: the molecule is produced by no organism. It belongs to the chemical class of diarylureas and is obtained by organic synthesis, with no connection to the plant cannabinoid pathway from CBGA.

Not classified in France. PSNCBAM-1 is not a tetrahydrocannabinol, nor an ester, an ether or a salt of THC: it is therefore not reached by the generic clause of annex IV of the decree of 22 February 1990, and it is explicitly named in no ANSM decree. Its status is that of a research reagent: transfer between laboratories is lawful, but the molecule holds no authorisation as a medicine, as a cosmetic ingredient or as a foodstuff, which rules out any use in a consumer product.
European Union
No classification at European Union level. The molecule does not appear among the substances placed under control under the European framework on new psychoactive substances, nor does it appear in the schedules of the international conventions of 1961 and 1971. It circulates solely as a chemical intended for research, subject to the general rules on chemical labelling and safety.

Diarylurée, hors de la famille des cannabinoïdes : squelette 1-(4-chlorophényl)-3-[3-(6-pyrrolidin-1-yl-pyridin-2-yl)phényl]urée, qui associe un phényle chloré, un pont urée et un biaryle pyridine-pyrrolidine. Aucun noyau résorcinol, aucune chaîne alkyle de type terpénophénolique.
Pharmacokinetics
Les données disponibles proviennent uniquement du rongeur. La molécule est très instable sur le plan métabolique, avec une demi-vie d'environ 13 minutes sur microsomes hépatiques de rat, ce qui a conduit les équipes à recourir à la voie intrapéritonéale et à des délais très courts entre l'injection et le test comportemental. Les effets centraux observés chez le rat et la souris impliquent un passage de la barrière hémato-encéphalique, mais aucune mesure de concentration cérébrale n'est publiée, et il n'existe aucune donnée humaine d'absorption, de distribution ou d'élimination.
Metabolism
La clairance hépatique rapide observée sur microsomes de rat oriente vers un métabolisme oxydatif de phase I, conforme à ce qu'on attend d'une diarylurée. Aucun métabolite n'a toutefois été isolé ni identifié dans la littérature accessible, et les enzymes responsables comme les éventuelles voies de conjugaison restent non documentées.
Toxicology and risks
Aucune étude de toxicité réglementaire, aucune donnée humaine. Chez la souris, les doses intrapéritonéales employées en pharmacologie expérimentale n'ont pas perturbé l'activité locomotrice, et les effets rapportés se limitent à une baisse de la prise alimentaire et des récompenses consommées. Le risque théorique reste celui de la classe : la réduction de la signalisation CB1 a conduit au retrait du rimonabant en 2008 pour effets dépressifs et anxieux, et rien ne permet aujourd'hui d'affirmer qu'une modulation allostérique négative en soit exempte. Cette molécule est un réactif de laboratoire et ne doit pas être ingérée.
Detection and analysis
Il n'existe aucune méthode de dépistage de routine. La molécule n'est intégrée ni aux panels toxicologiques cliniques, ni aux criblages de nouvelles substances psychoactives, faute de circulation sur le marché récréatif. Une identification supposerait une méthode ciblée en chromatographie liquide couplée à la spectrométrie de masse, construite à partir d'un étalon de référence commercial.
