In plain terms
SR144528 is an entirely artificial molecule, developed in the Sanofi group's laboratories in the late 1990s. It is not found in hemp and does not reproduce the effects of cannabis: on the contrary, it blocks one of the two receptors on which cannabinoids act, the one mainly present in immune cells. It serves solely as a laboratory tool and has never been administered to patients.

Pharmacology
SR144528 is the first potent and selective antagonist of the CB2 cannabinoid receptor, described in 1998 by the teams at Sanofi Recherche. Its affinity for CB2 is subnanomolar whereas its affinity for CB1 is about 700 times lower, a selectivity that made it the reference tool for attributing, or not, an observed effect to the CB2 receptor. It blocks at nanomolar concentrations the responses induced by CB2 agonists, namely inhibition of adenylyl cyclase, activation of the p42 and p44 MAP kinases and B-lymphocyte activation, without touching the equivalent CB1-dependent pathway. The literature has also established that it is not a simple blocker: SR144528 behaves as an inverse agonist, reversing the constitutive activity of CB2 and acting on type V and type II adenylyl cyclases through a Gi protein-dependent mechanism. What research subsequently brought to light qualifies its status as a clean tool: the molecule inhibits acyl-CoA cholesterol acyltransferase independently of CB2, and exerts inverse agonist activity at the orphan receptor GPR6, two off-target actions that may contribute to effects attributed to CB2 blockade. No human data exist, either on pharmacokinetics or on tolerability: the molecule has never left the laboratory and its metabolism has not been characterised.

Origin (research tool)
No natural biosynthetic pathway. SR144528 is a product of organic synthesis in the diarylpyrazole class, obtained by building the diarylated pyrazole core and then forming a carboxamide bearing a fenchyl group. The molecule has never been isolated from a plant, a fungus or an animal tissue, and no known enzyme produces it.
Legal framework
France
SR144528 is not listed by name on any French list of narcotics or psychotropic substances. Nor is it caught by the generic clause of annex IV of the decree of 22 February 1990, which covers only tetrahydrocannabinols, their esters, ethers and salts, as well as the salts of the aforementioned derivatives: SR144528 is a synthetic diarylpyrazole, with no structural link to THC and no cannabinoid agonist-type effect. The successive generic classifications of families of synthetic cannabinoids decided by the ANSM cover compounds that reproduce the action of delta-9-THC at the receptors, which is not the case for a CB2 antagonist. The situation is therefore that of an unclassified substance. This does not mean it may be marketed: no marketing authorisation exists, and the molecule has no legal use in a consumer product, a food supplement or a cosmetic. It circulates only as a laboratory reagent.
European Union
No classification at European Union level. SR144528 has never obtained a marketing authorisation from the European Medicines Agency and does not figure among the new psychoactive substances subject to control measures under Framework Decision 2004/757/JHA as amended. Nor is it listed in the schedules of the United Nations conventions of 1961 and 1971. Its marketing is confined to the market for research reagents, governed by chemicals regulation and not by narcotics law.

Pharmacokinetics
Les seules données disponibles proviennent de l'animal. Administré par voie orale chez la souris, SR144528 déplace la liaison d'un radioligand sur les récepteurs CB2 de la rate à faible dose et avec une durée d'action longue, alors qu'il ne déplace pas la liaison sur les récepteurs CB1 du cerveau par cette même voie : la distribution est donc essentiellement périphérique. Aucune donnée de pharmacocinétique humaine n'existe, la molécule n'ayant jamais été administrée à l'homme dans un essai publié, et ni la biodisponibilité, ni le volume de distribution, ni la demi-vie ne sont documentés chez l'espèce humaine.
Metabolism
Le métabolisme de SR144528 n'a pas fait l'objet d'une caractérisation publiée : les voies d'oxydation, les isoenzymes du cytochrome P450 impliquées et les métabolites formés ne sont pas décrits dans la littérature accessible. Les travaux disponibles portent sur la pharmacologie du récepteur et n'abordent pas le devenir de la molécule dans l'organisme.
Toxicology and risks
Les données humaines manquent totalement : SR144528 n'a jamais été évalué chez l'homme, aucun essai clinique ne lui est consacré et aucune étude de toxicité chronique n'a été publiée. Sur cellules et chez l'animal, le principal point de vigilance est une activité hors cible : la molécule inhibe l'acyl-CoA cholestérol acyltransférase, aussi appelée stérol O-acyltransférase, et réduit la synthèse d'esters de cholestérol dans plusieurs modèles de macrophages ainsi que dans les microsomes hépatiques de souris, y compris en l'absence de récepteur CB2. Cette action complique l'interprétation des expériences qui attribuent au seul blocage du CB2 les effets observés, et elle constitue en soi un effet pharmacologique non anticipé sur le métabolisme lipidique. Une action d'agoniste inverse sur le récepteur orphelin GPR6 a également été rapportée. Le produit est distribué comme réactif de laboratoire avec une mention d'usage non humain, et aucun usage en produit de consommation n'est documenté ni défendable en l'état des connaissances.
Detection and analysis
Aucune méthode de dépistage de routine ne vise SR144528. La molécule ne figure pas dans les panels toxicologiques usuels, ni parmi les substances suivies par le système d'alerte précoce européen sur les nouvelles substances psychoactives, et la littérature ne rapporte aucun cas médico-légal impliquant ce composé, ce qui s'explique par l'absence d'effet de type cannabique et donc d'usage récréatif. Dans les travaux de recherche, l'identification et le dosage reposent sur la chromatographie liquide couplée à la spectrométrie de masse, à partir d'un étalon analytique commercial.
