Pharmacology
SSR411298 belongs to the family of reversible inhibitors of FAAH, the enzyme responsible for the hydrolysis of anandamide. The inhibition is competitive and genuinely reversible, as dialysis experiments confirm, with a median inhibitory concentration of 62.5 nM on mouse brain; by the oral route, the dose inhibiting half of brain enzyme activity is 0.17 mg/kg in mice. The result is a marked rise in anandamide, oleoylethanolamide and palmitoylethanolamide in the hippocampus, with no change in 2-AG. The molecule has no direct affinity for CB1 or CB2 up to 10 µM: it acts solely by letting the endogenous mediators accumulate. In rodents, it does not reproduce the profile of direct cannabinoid agonists, with neither catalepsy, nor ataxia, nor substitution for WIN 55,212-2, and it does not precipitate withdrawal on rimonabant challenge. Animal models showed antidepressant-like activity and an anxiolytic effect restricted to highly aversive situations. Clinically, the picture is quite different: a phase 2 trial conducted by Sanofi in 527 elderly patients with major depression ended in 2010 without any result being filed in the registry or published in a peer-reviewed journal, and a second phase 2 trial in persistent cancer pain was stopped in 2012 after five inclusions, for strategic reasons. Human data are therefore almost entirely lacking, and no therapeutic benefit has been established for this molecule.

Origin (research tool)
No biological pathway produces this molecule. It is obtained by organic synthesis from a methoxylated naphthalene intermediate, the glycinamide carbamate motif being installed at the end of the sequence.

SSR411298 is not a tetrahydrocannabinol and is neither an ester, an ether nor a salt of one: it therefore escapes the generic clause of the decree of 22 February 1990, annex IV, which covers tetrahydrocannabinols, their esters, ethers, salts as well as the salts of the aforementioned derivatives. Nor is it listed by name on the lists of narcotics or psychotropics maintained by ANSM. It is therefore not classified as a narcotic in France. That does not make it available for all that: it is an abandoned drug candidate, devoid of any marketing authorisation, so that any sale intended for human, food or cosmetic use would be unlawful. Its place is the laboratory, not a consumer product, and it enters into no CBD-based preparation.
European Union
No specific control at European Union level. The substance does not figure in the schedules of the United Nations conventions of 1961 and 1971, has been the subject of no European control decision and is not recorded by the EUDA (formerly EMCDDA) among the new psychoactive substances circulating on the market. It is available only as a research reagent, a status that authorises neither human consumption nor marketing to the public in the Member States.

Pharmacokinetics
Actif par voie orale et pénétrant dans le cerveau chez le rongeur. L'inhibition de la FAAH cérébrale mesurée ex vivo chez la souris apparaît dès 0,17 mg/kg par voie orale, devient maximale au-delà de 1 mg/kg, culmine entre 60 et 360 minutes, se maintient environ six heures puis décroît régulièrement pour rejoindre le niveau de base en vingt-quatre heures. Chez l'humain, les essais cliniques ont employé des prises orales quotidiennes de 10, 50 et 200 mg, mais aucun paramètre pharmacocinétique humain, concentration maximale, demi-vie ou exposition, n'a été publié.
Metabolism
Non documenté dans la littérature publique : ni les métabolites, ni les enzymes hépatiques impliquées, ni la voie d'élimination n'ont été décrits pour cette molécule. La fonction carbamate constitue le point chimiquement labile de la structure, mais aucune étude publiée n'en précise le devenir chez l'animal ou chez l'humain.
Toxicology and risks
Chez le rongeur, la molécule ne reproduit pas la tétrade cannabinoïde : pas de catalepsie ni d'ataxie jusqu'à 100 mg/kg, pas de substitution au WIN 55,212-2 jusqu'à 500 mg/kg par voie orale, seules une légère hypothermie et une action antinociceptive ont été relevées, et le rimonabant ne provoque pas de sevrage après administration répétée. Chez l'humain, les auteurs évoquent une douzaine d'études de tolérance en dose unique ou répétée jugées favorables, mais ces données sont citées comme communication personnelle et n'ont jamais été publiées ; aucun essai n'a déposé de résultats de sécurité au registre. Le contexte de la classe reste lourd, l'inhibiteur irréversible BIA 10-2474 ayant provoqué des atteintes neurologiques graves lors d'un essai de phase 1 conduit à Rennes en 2016. Le caractère réversible du SSR411298 est présenté comme un argument de sécurité, mais cette hypothèse n'a jamais été démontrée en clinique, et aucune donnée de toxicité chronique humaine n'existe.

La substance n'est recherchée ni par les immunoessais urinaires classiques ni par les panels toxicologiques hospitaliers, et aucune méthode analytique validée ni aucun métabolite marqueur n'a été publié dans la littérature médico-légale. Une identification supposerait une chromatographie liquide couplée à la spectrométrie de masse, avec un étalon de référence dédié.
