Michael Walker et de Vincenzo Di Marzo, lors de l'analyse approfondie de l'extrait de striatum bovin dont le NADA avait été isolé. In plain terms
N-stearoyl dopamine is a small fatty molecule that the mammalian body makes itself, by assembling a common fatty acid and dopamine. It does not come from hemp and is present in no product sold in a shop. Taken alone, it triggers nothing in the published assays: it merely reinforces the action of other, related molecules on a sensory sensor, and in humans the data are lacking.

Pharmacology
N-stearoyl dopamine belongs to the N-acyl dopamines, the same family as NADA, but its saturated eighteen-carbon fatty chain changes everything. On identifying it in the bovine striatum in 2003, Chu and colleagues found that it does not activate the TRPV1 channel, that it provokes no pain behaviour in the rat and that it does not displace the CB1 receptor radioligand below 5 µM: the long unsaturated chain is missing, and with it the direct activity. Its interest lies elsewhere. De Petrocellis and colleagues showed in 2004 that pre-incubation with this molecule lowers the median effective concentration of NADA at human TRPV1 from about 90 nM to about 30 nM, amplifies the response to anandamide and acts synergistically with acidification of the medium. Co-injected with NADA into the rat hind paw, it accentuates thermal hyperalgesia, and it increases carrageenan-induced nociceptive behaviour. This is the classic profile of a so-called entourage compound: inactive in itself, it alters the sensitivity of the target to active ligands. Beyond these two studies and an isolated report on its hepatic sulfation in the rat, the literature is very thin: human data are lacking, no physiological role is established and no therapeutic indication can be claimed.

Biosynthetic pathway (in vivo)
The route of formation is not established. By analogy with the other N-acyl dopamines, an enzymatic condensation between dopamine and stearic acid, or its activated derivative, is the hypothesis retained. The enzyme PM20D1, identified as capable of condensing dopamine and arachidonic acid into NADA, has not been studied with the stearic substrate. No synthetic enzyme has therefore been named for this molecule, and its tissue levels have never been followed after pharmacological manipulation.
Legal framework
France
N-stearoyl dopamine is a lipid produced by the mammalian body. It appears by name on no ANSM list of narcotics or psychotropics, and it does not fall within the generic clause of annex IV of the decree of 22 February 1990, which covers tetrahydrocannabinols, their esters, their ethers, their salts as well as the salts of the aforementioned derivatives: the molecule is neither a tetrahydrocannabinol nor a tetrahydrocannabinol derivative, but an amide of a saturated fatty acid and a catecholamine. It is therefore not classified as a narcotic in France. Its only documented circulation is that of a laboratory reagent and an analytical standard, with no food, cosmetic or medicinal status.
European Union
No Member State schedules N-stearoyl dopamine under the international conventions of 1961 and 1971 on narcotic drugs and psychotropic substances, and the molecule is the subject of no notification to the European Union early warning system on new psychoactive substances, whose scope covers products appearing on the recreational market. An endogenous compound used as a laboratory tool, it remains outside the perimeter of Framework Decision 2004/757/JHA. No marketing authorisation as a medicine and no assessment under the novel foods regulation concern it.

N-acyl-dopamine saturée : liaison amide entre l'acide stéarique, acide gras saturé à dix-huit carbones, et la dopamine. La molécule associe donc une longue chaîne aliphatique dépourvue de double liaison à un noyau catéchol. Elle relève de l'endocannabinoïdome élargi et non des cannabinoïdes au sens structural du terme : ni noyau terpénophénolique, ni cycle résorcinol. Par rapport au NADA, la seule différence tient à la chaîne grasse, saturée ici et tétra-insaturée chez le NADA, et cette différence suffit à supprimer l'activité directe sur les cibles connues.
Pharmacokinetics
Aucun paramètre pharmacocinétique n'a été établi, ni chez l'être humain ni chez l'animal : la molécule n'a jamais fait l'objet d'une administration systémique documentée. Les seules expositions in vivo publiées sont des injections locales dans la patte postérieure du rat, à des doses de l'ordre du microgramme, destinées à tester son effet sur la réponse au NADA. Comme les autres amides d'acides gras à longue chaîne, elle est très lipophile et s'associe fortement aux membranes cellulaires, ce qui limite sa diffusion libre dans les compartiments aqueux.
Metabolism
Une seule voie a été mesurée directement, la sulfatation. Dans des fractions cytosoliques de tissus de rat, les N-acyl-dopamines sont conjuguées par des sulfotransférases, et la N-stéaroyl-dopamine n'a été sulfatée que dans le foie, alors que ses congénères insaturés le sont aussi dans le système nerveux. Les auteurs décrivent cette voie comme la plus productive de la famille, tout en soulignant qu'une constante de Michaelis élevée en rend la portée physiologique incertaine. L'hydrolyse de la liaison amide par la FAAH et la méthylation du noyau catéchol par la COMT sont attendues par analogie avec le NADA, mais elles n'ont pas été démontrées sur cette molécule.
Toxicology and risks
Aucune donnée de toxicité humaine n'existe, faute d'exposition documentée. Chez le rat, l'injection locale ne produit ni hyperalgésie thermique ni comportement nociceptif lorsqu'elle est administrée seule. Le seul signal in vivo est une majoration de la douleur provoquée par le NADA et par la carragénine, ce qui traduit une amplification de signalisation plutôt qu'une toxicité propre. Aucune évaluation réglementaire de sécurité, aucune étude de toxicité par administration répétée et aucun essai clinique ne portent sur cette molécule, si bien qu'aucun usage humain ne peut être envisagé.
Detection and analysis
L'identification initiale dans le tissu nerveux bovin repose sur la spectrométrie de masse à temps de vol couplée à un quadripôle, appliquée à des extraits lipidiques. Pour les matrices biologiques humaines, la molécule figure parmi les analytes d'une méthode de chromatographie liquide couplée à la spectrométrie de masse en tandem, en mode de suivi de réactions multiples, avec extraction en phase solide en ligne sur sérum et des limites de quantification de l'ordre du picogramme par millilitre ; aucune valeur de concentration sérique n'a toutefois été publiée. Étant endogène et non classée, la molécule ne figure dans aucun criblage toxicologique de routine.
