1972 : Vree, Breimer, van Ginneken et van Rossum identifient par chromatographie en phase gazeuse couplée à la spectrométrie de masse un homologue du Δ9-THC à chaîne méthyle dans un extrait de cannabis brésilien, aux côtés des équivalents C1 du CBD et du CBN. Harvey signale en 1976 la forme acide correspondante, l'acide tétrahydrocannabiorcolique. Ross et ElSohly le retrouvent en 2005 parmi les seize cannabinoïdes du pollen de plants mâles de Cannabis sativa. Sa pharmacologie n'est décrite qu'en 2011, par l'équipe d'Andersson, Bevan, Högestätt et Zygmunt.

Pharmacology

Tetrahydrocannabiorcol (Δ9-THC-C1) is the methyl homologue of Δ9-THC: the same benzo[c]chromene skeleton of (6aR,10aR) configuration, but a side chain of a single carbon at position 3. This truncation takes it outside the classical cannabinoid activity domain, which requires a chain of five to eight carbons. The study published in Nature Communications in 2011 describes it as devoid of agonist properties at the human CB1 and CB2 receptors, and identifies its true target: the TRPA1 ion channel. At human TRPA1 expressed in HEK293 cells, it proved the most active of the cannabinoids and derivatives screened, as potent and as effective as allyl isothiocyanate, through a non-electrophilic mechanism. In cultured sensory neurons, a concentration of 20 µM triggered a calcium response in 38 % of neurons from wild-type mice against 3 % in mice lacking TRPA1, and intrathecal injection produced antinociception that was absent in those same mice. The demonstration stops there: no trial in humans, and no pharmacokinetic, metabolic or toxicological data have been published, while the concentrations active in vitro lie in the micromolar range. This molecule should therefore be read as a pharmacological tool serving to dissect spinal TRPA1, not as a cannabinoid for consumption.

Biosynthetic pathway

The methyl homologue of Δ9-THC: the pentyl chain carried at position 3 of the benzo[c]chromene skeleton is here reduced to a simple methyl group. The orcinoid (C1) series is attached by analogy to a short polyketide precursor of the orsellinic acid type, prenylated and then cyclised as in the pathway leading to THC; the enzymology specific to this short series has never been demonstrated experimentally. The acid form, tetrahydrocannabiorcolic acid, has also been detected in cannabis, which remains consistent with a biosynthetic rather than an artefactual origin.

Cannabinoïde classique de type dibenzopyrane (squelette benzo[c]chromène), configuration trans (6aR,10aR) identique à celle du Δ9-THC, avec la double liaison en position 9. Il appartient à la série dite orcinoïde : la chaîne latérale en position 3 se réduit à un méthyle, contre propyle pour le THCV, butyle pour le THCB, pentyle pour le THC et heptyle pour le THCP. Ses homologues de série, décrits dans les mêmes travaux de 1972, sont le cannabidiorcol (CBD-C1) et le cannabiorcol (CBN-C1).

Pharmacokinetics

Aucune donnée de pharmacocinétique n'est publiée, ni chez l'humain ni chez l'animal : biodisponibilité, délai d'action, distribution tissulaire et demi-vie restent inconnus. Les seules administrations documentées sont expérimentales et intrathécales chez la souris, une voie qui contourne entièrement l'absorption et la distribution systémique et qui ne permet donc aucune extrapolation vers une prise orale ou inhalée. Le raccourcissement de la chaîne latérale abaisse la lipophilie par rapport au THC, mais aucune mesure de passage de la barrière hématoencéphalique n'a été rapportée.

Metabolism

Aucune voie métabolique n'a été caractérisée pour ce composé : ni enzymes de phase I (cytochromes P450), ni conjugaisons de phase II, ni métabolite actif identifié. Par analogie avec le Δ9-THC, une hydroxylation allylique en position 11 suivie d'une oxydation en acide carboxylique serait plausible, mais aucune étude ne l'a établie pour la série à chaîne méthyle.

Toxicology and risks

Aucune étude de toxicologie n'existe pour le tétrahydrocannabiorcol : pas de dose sans effet observé, pas de données d'exposition répétée, pas de cas clinique publié. L'absence d'activité agoniste CB1 laisse penser qu'il ne reproduit pas les effets aigus du THC, mais cette absence d'intoxication cannabinoïde ne vaut pas démonstration d'innocuité. Le TRPA1 est le capteur des stimuli chimiques nocifs et un médiateur de l'irritation cutanée et viscérale : activer ce canal constitue en soi un signal à surveiller, l'effet antinociceptif observé chez la souris dépendant étroitement du site d'administration. Les données humaines manquent.

Detection and analysis

La molécule a toujours été mise en évidence par chromatographie en phase gazeuse couplée à la spectrométrie de masse, avec un ion moléculaire à m/z 258, soit 56 unités de masse de moins que le Δ9-THC : c'est précisément cet écart qui a permis son identification dans le haschich et le cannabis en 1972, puis dans le pollen de plants mâles en 2005. Aucune méthode validée de dosage en matrice biologique n'a été publiée, et les immunoessais urinaires courants, calibrés sur le THC-COOH, ne la recherchent pas. En pratique, elle n'apparaît que dans les profils cannabinoïdes complets établis en laboratoire de recherche.
