WOBE437 is a laboratory molecule, developed by Swiss chemists from a substance present in pepper. It does not come from hemp and enters into no product on sale. Pharmacology
WOBE437 is the lead compound of a class of pharmacological tools called selective endocannabinoid reuptake inhibitors. Instead of activating the cannabinoid receptors, it is thought to slow the entry of anandamide and 2-arachidonoylglycerol into cells, which locally prolongs their signal without touching the degradative enzymes FAAH and MAGL. The original paper reports an IC50 of around 10 nM on anandamide uptake and an absence of direct affinity for CB1 as for CB2. In the mouse, the molecule is orally bioavailable, reaches the brain in under twenty minutes and is cleared from it in about three hours; the effects described, a reduction of anxiety, of inflammatory pain and of the severity of an autoimmune encephalomyelitis, disappear in the presence of antagonists of CB1, CB2, TRPV1 or PPARγ depending on the protocol. This picture must be read with caution: an independent team demonstrated in 2022 three off-target proteins, SCCPDH, VAT1 and ferrochelatase, and observed in neuroblastoma culture a modulation of uptake the inverse of that expected. The postulated endocannabinoid transporter has still not been isolated, no clinical trial has been conducted and human data are entirely lacking.

Origin (research tool)
No known biological pathway: WOBE437 is produced by no living organism. It is obtained chemically, by formation of an amide bond between a twelve-carbon dienoic acid and 3,4-dimethoxyphenethylamine. The starting skeleton, that of the pepper alkamides, is the only link with the plant world.
Legal framework

WOBE437 is named in no French decree and appears on no ANSM list. Nor is it caught by the generic clause of annex IV of the decree of 22 February 1990, which covers the tetrahydrocannabinols, their esters, their ethers, their salts as well as the salts of those derivatives: this molecule is a synthetic alkamide, devoid of a tetrahydrocannabinol skeleton. Its status is therefore that of an unscheduled substance. That does not make it marketable, however: it holds no marketing authorisation as a medicinal product and is admitted neither as a food supplement nor in cosmetics. Its actual use is confined to the laboratory.
European Union
No European Union text classifies WOBE437. It does not figure among the new psychoactive substances monitored by the EUDA (formerly the EMCDDA) under Regulation 2017/2101, and appears in no schedule of the United Nations conventions of 1961 and 1971. Nor does it have novel food status. It circulates solely as a reagent intended for research.

Alcamide de synthèse de type N-(3,4-diméthoxyphénéthyl)dodéca-2,4-diénamide, apparentée aux pipéramides du poivre. Ce n'est pas un cannabinoïde au sens structural : la molécule ne possède ni noyau résorcinol, ni chaîne alkyle latérale, ni cycle terpénique caractéristiques des phytocannabinoïdes. Elle se rapproche davantage des amides d'acides gras, ce qui explique sa forte lipophilie.
Pharmacokinetics
Chez la souris, WOBE437 est biodisponible par voie orale : le pic plasmatique et le pic cérébral sont atteints en vingt minutes ou moins, avec environ 470 pmol/g dans le cerveau après une dose de 50 mg/kg, soit une concentration cérébrale estimée proche de 400 nM. Le volume de distribution apparent est très élevé, ce qui reflète une lipophilie marquée et une rétention tissulaire importante. Les concentrations les plus fortes se retrouvent dans le foie, puis le rein et la rate. La molécule est éliminée du cerveau en environ 180 minutes. Aucune donnée de pharmacocinétique humaine n'a été publiée.
Metabolism
Le devenir métabolique n'a pas été caractérisé en détail. Des essais sur microsomes hépatiques humains et murins mettent en évidence une clairance intrinsèque mesurable, plus rapide avec les microsomes humains qu'avec les microsomes de souris, ce qui oriente vers une prise en charge hépatique oxydative. La fonction amide et les deux groupements méthoxy du noyau aromatique sont les points de transformation attendus, mais aucun métabolite n'a été formellement identifié dans la littérature publiée.
Toxicology and risks
Aucune étude de toxicologie réglementaire n'a été publiée. Dans les modèles murins, des administrations répétées de 10 mg/kg pendant vingt jours ont été tolérées, sans catalepsie, sans altération notable de la locomotion, sans variation significative de la température corporelle et sans désensibilisation du récepteur CB1. Ces observations ne constituent pas une évaluation de sécurité : il n'existe ni dose sans effet établie, ni donnée d'exposition humaine, ni étude de génotoxicité, ni étude de toxicité pour la reproduction. L'identification de la ferrochélatase parmi les protéines liées invite à la prudence, cette enzyme intervenant dans la dernière étape de la synthèse de l'hème.
Detection and analysis
Aucune méthode de dépistage de routine n'existe et la molécule n'est recherchée dans aucun panel toxicologique courant, urinaire ou sanguin. Les travaux publiés la quantifient dans le plasma et les tissus par chromatographie liquide couplée à la spectrométrie de masse en tandem, avec étalons internes. Elle n'a fait l'objet d'aucun signalement au système d'alerte précoce européen sur les nouvelles substances psychoactives, ce qui reflète l'absence de circulation hors du cadre académique.
