- Molécule issue des laboratoires Shionogi, au Japon. La série chimique et l'identification du composé ont été publiées en 2012 par Odan et coll. dans Bioorganic & Medicinal Chemistry Letters, au terme d'une optimisation des relations structure-activité autour du motif 3-carbamoyl-2-pyridone. Le développement clinique avait en réalité commencé plus tôt : deux essais chez des patients atteints de dermatite atopique ont été enregistrés par Shionogi et conduits entre 2007 et 2009. La caractérisation pharmacologique détaillée a suivi entre 2014 et 2017, puis une équipe universitaire néo-zélandaise a repris la molécule en 2023 comme point de départ chimique sous le nom de code UOSS77.

In plain terms
S-777469 is a laboratory molecule, developed in the 2000s by a Japanese pharmaceutical company. It has nothing to do with hemp: it was purpose-built to bind to just one of the body's two cannabinoid receptors, the one found mainly in immune cells and in the skin. Studied against the itching of eczema, it was abandoned before reaching the pharmacy.

Pharmacology
S-777469 belongs to the 3-carbamoyl-2-pyridone chemical family and shares no structure with the plant cannabinoids. Its interest lies in its selectivity: it activates the CB2 receptor with an affinity measured at a pKi of 6.73 at the human receptor, whereas it displaces the ligands of the CB1 receptor only very partially, which in principle rules out the central effects associated with THC. On the signalling pathways, it acts as a full agonist on cyclic AMP inhibition and as a partial agonist on beta-arrestin 2 recruitment, with no measurable bias between the two. In rodents, oral administration dose-dependently reduces scratching provoked by compound 48/80, by histamine, by substance P or by serotonin, an effect abolished by the CB2 antagonist SR144528; the molecule also inhibits histamine-induced peripheral nerve firing, reduces epidermal thickness and limits the infiltration of mast cells and eosinophils in dermatitis models. That is where the demonstration stops. In humans, the substance is well absorbed orally and extensively metabolised, but the two clinical trials conducted in patients with atopic dermatitis between 2007 and 2009 gave rise to no posting of results on the public register, and the programme was discontinued. No therapeutic benefit is established in humans, and human medium- and long-term safety data are lacking.

Origin (research tool)
No known biological route: the molecule is entirely synthetic and has no natural precursor. Its 3-carbamoyl-2-pyridone skeleton belongs to pharmaceutical synthetic chemistry and lies outside the cannabigerol pathway that gives rise to the hemp cannabinoids.

France
S-777469 is neither a tetrahydrocannabinol nor an ester, ether or salt of THC: it is therefore not caught by the generic clause of annex IV of the decree of 22 February 1990. Nor is it listed by name on the narcotics or psychotropics lists maintained by the ANSM. The molecule is therefore not classified in France. This absence of classification does not amount to authorisation: it is an experimental substance with no marketing authorisation, whose development has been halted. Its sale, or its incorporation into a consumer product, a food supplement or a cosmetic, would fall under medicines and novel food regulation, and would on that basis be unlawful.
European Union
No international classification is documented: the molecule appears neither in the schedules of the 1961 Single Convention nor in those of the 1971 Convention, and no WHO scheduling recommendation has been published concerning it. It has not been notified as a new psychoactive substance on the European market monitored by the EUDA, which is explained by its lack of effect at CB1 and hence of any sought-after psychoactive property. Its status in the European Union remains that of an abandoned drug candidate: use restricted to research, with no food or cosmetic authorisation.

Dérivé 3-carbamoyl-2-pyridone, c'est à dire un 2-oxo-1,2-dihydropyridine-3-carboxamide portant un groupe 4-fluorobenzyle sur l'azote et un acide cyclohexane-1-carboxylique sur l'amide. Ligand CB2 dit non cannabinoïde : aucune parenté avec le squelette terpénophénolique des cannabinoïdes du chanvre, ni avec les structures indoliques ou indazoliques des cannabinoïdes de synthèse récréatifs.
Pharmacokinetics
Composé actif par voie orale. Chez le rat et chez le chien, la biodisponibilité mesurée se situe entre 50 et 70 %, avec une excrétion principalement biliaire puis fécale, achevée pour l'essentiel en moins de 48 heures. Chez des volontaires sains ayant reçu une dose orale marquée au carbone 14, la radioactivité totale est absorbée rapidement et le pic plasmatique survient vers la quatrième heure. La molécule inchangée ne représente qu'environ 30 % de la radioactivité circulante, ce qui traduit une transformation métabolique importante. L'élimination se fait majoritairement par les fèces.
Metabolism
Le métabolisme est extensif et bien documenté chez l'animal comme chez l'humain. Les deux principaux métabolites circulants chez l'humain sont le dérivé 5-carboxylique et le dérivé 5-hydroxyméthyle ; un glucuronide d'acyle et diverses formes hydroxylées sont retrouvés chez le rat et chez le chien, sans qu'aucun métabolite propre à l'espèce humaine n'ait été identifié. La formation des deux métabolites oxydés est catalysée par le cytochrome CYP2C9. Une divergence notable a été décrite entre les essais sur hépatocytes ou microsomes et l'observation clinique : les acides gras à longue chaîne, dont l'acide arachidonique, l'acide alpha-linolénique, l'acide oléique et l'acide myristique, inhibent l'oxydation par le CYP2C9, tandis que l'albumine sérique la favorise, ce qui explique la sous-estimation de la voie oxydative in vitro.
Toxicology and risks
Les données publiées restent limitées. Les travaux non cliniques ont conclu que le rat et le chien constituaient des espèces pertinentes pour l'évaluation toxicologique, mais les rapports de toxicité eux-mêmes n'ont pas été rendus publics dans la littérature accessible. Deux essais cliniques chez des patients atteints de dermatite atopique, dont un mené sur douze semaines auprès de plus de deux cents participants, comportaient la tolérance parmi leurs critères d'évaluation ; aucun résultat n'a été déposé sur le registre public et le programme a été arrêté. En l'absence d'activité significative sur CB1, aucun effet psychoactif n'est attendu, mais les données humaines de sécurité manquent et la molécule n'a jamais été autorisée comme médicament. La dépendance de son métabolisme oxydatif au seul CYP2C9 laisse par ailleurs présager un potentiel d'interactions médicamenteuses qui n'a pas été caractérisé publiquement.
Detection and analysis
La molécule n'est pas recherchée en toxicologie analytique de routine et ne figure pas parmi les cannabinoïdes de synthèse surveillés sur le marché des nouvelles substances psychoactives, faute d'usage récréatif documenté. En contexte expérimental, elle est dosée dans les matrices biologiques par chromatographie liquide couplée à la spectrométrie de masse en tandem. Une méthode d'imagerie par spectrométrie de masse MALDI, appuyée sur des étalons préparés en tissu de substitution, a été publiée pour cartographier sa distribution dans les organes de la souris, avec une bonne concordance entre les deux approches.
